Objective To research the feasibility、 modeling standards, and successcriteria in modeling of opening-method to establish the rat model of endometriosis,and explore the effects of estrogen in transplanting-method to establish the ratendometriosis model.Methods①Modeling:37rats were obtained,each rat was operated withopening-method and transplanting-method. Through microscopic view of thenaked eye and view of the endometrial tissue HE staining after endoscopic tocompare the characteristics of the modeling method.②Expression of MMP-9:Detected the expressions of MMP-9with immunohistochemical method in eutopicand ectopic tissue.③Estrogen intervention:37rats were randomly divided intofour groups:Group A:Giving estrogen before making models;Group B:Givingestrogen after making models;Group C:Giving estrogen during the progress ofmaking models; Group D: The normal control group. Using transplanting-methodto build the endometriosis models in rat, Observe the characteristics of eachmodel,Compared the achievement of ratio、 diameter of rat uterus、 the size ofectopic cyst and the intensity of the expression of MMP-9in models.Results①The model achievement ratio of opening-method is78.37%,there was no difference with transplanting-method.②Cystic mass is mainclinical signs with81.08%in transplanting-method,followed by flame shape.40.54%clinical signs was flame shape in opening-method,followed by darkblue nodules. The numbers of ectopic lesions and surrounding tissue adhesion intransfer method was significantly lower then Opening-method(P<0.05).Therewere six cases of intestinal obstruction in Opening-method, which significantlyhigher then transplanting-method(P<0.05).③The achievement ratio of modingwas no difference in four groups.④The view of naked eye was similarin four groups,There was7ectopic lesions adhesioned with thesurrounding tissue in group C,which was significantly more than threeother group, and there was no difference in the other groups.⑤TheUterine diameter of group A(0.47±0.035cm)was significantly large than groupD(0.34±0.033cm) before making models. The uterine diameter after modelcompared with modeling were significantly increased in four groups of rats,therewas statistically significant difference. When second operation: the uterinediameter of group D was significantly small then other groups, And the uterinediameter in group C were significantly large then other groups,the uterinediameter of group B was significantly large then group A.⑥Compared to group Band D,the volume of cysts in group C were significantly big, but showed the samevolume in group A, the volume of cysts in group B and group D was no difference.⑦The MMP-9expressed in eutopic and ectopic tissues, and the expression ofMMP-9in ectopic endometrium was stronger than eutopic endometrium. Theexpression of MMP-9in group C was significantly higher than group A、 B andD,which showed the same expression.Conclusions①The two models both have high achievement ratios,Opening-method to establish the EMS model in rats was feasible.②The twomodels have different features, the opening-method model,s general view wasmore similar to human endometriosis performance, it has extensive lesions, large area,simple、feasible and other major features.③The opening-method model hasmuch complications, lesions dispersion. It,s complications, diffuse lesions, thematerial is relatively difficult, damage to rat is great, which is unfavorable to thefollow-up of drug testing.④Estrogen had no effect on the success rate ofmodeling.⑤Estrogen had no effect on the general clinical manifestations of thefilm, but can influence the complications.⑥Estrogen can reduce the operativedifficulty of transplantation modeling, and influence uterus diameter、 cysts sizeand the expression of MMP-9.
