Objective:To observe the effect of Tianbing Tiaodu Capsule on rat with stagnation of waterconsumption,food intake, syrup preference,emotional behaviors and spatial memorycapacity,as well as the gastric antrum and plasma,hypothalamus somatostatin (SS) contentinfluences,from neuroendocrine and brain gut peptides reveal the essence of syndrome ofstagnation.Methods:60Wistar rats were randomly divided into6groups,each group of10: the normalgroup,model group,Chaihu Shugan powder group, deanxit and domperidone group,TianbingTiaodu capsule low dose group, Tianbing Tiaodu capsule high dose group.Rats in each groupwere adaptive feeding a week.To adopt the" tail clamping angered law" making rat model ofliver depression syndrome. After injecting,the normal group and model group rats daily tovolume15ml/kg Physiological saline by gavage,other groups gavage volume was15ml/kg,according to people and animal equivalent doses were converted into rat doseequivalent dosage:Chaihushugansan group will Chaihushugansan original Decoctionconcentrated,made of17.5g/(kg·d) concentrate, with combined Motilium group with10%Arabia rubber suspension prepared into medicine concentration16mg/(kg·d) of Deanxit anddrug concentration of6mg/(kg·d) of domperidone suspension. Tianbing Tiaodu capsule with10%Arabia rubber match drug concentrations were40mg/(kg·d) low dose group and80mg/(kg·d) of high dose group of suspension,a total of gavage daily,was administered fortwo weeks.Time for every rat prior quantitative good two bottles of water:1bottles of1%sucrose,1bottles of water, sugar and water consumption was observed in24h,sugarpreference=sugar consumption/total consumption of liquid x100%.Daily rats were given300g mouse grain,in the measurement of residual volume.After one week of treatment,all ratsin order to open field test and observation of trapping reaction,step-down test and Morriswater maze experiment. After one week,each group of fasted rats24h executed,byradioimmunoassay for measurement of rat plasma,gastric antrum and SS content inhypothalamus. Results:1Rats drinking volume:molding period,in addition to the normal group the amount ofwater is relatively stable,the rest of the stimulation group water volume decreasedsignificantly,compared with the normal group were significantly different (P <0.05).Aftertreatment,the treatment group compared with the model group,with significant difference(P<0.05),the days of high dose group and other treatment groups had significant difference(P<0.05),with statistical significance.Sugar preference:model group compared with normalgroup,sugar preference was significantly reduced,the difference was statistically significant(P<0.05).The treatment group compared with the model group,had significant difference (P <0.05).2Food-intake: molding period, in addition to the normal set of intake is more stable, therest of the stimulation group intake decreased significantly compared with the normalgroup,there were significant differences (P<0.05),with statistical significance.Aftertreatment,the treatment group compared with the model group,with significant difference(P<0.05),the days of high dose group and other treatment groups had significant difference(P<0.05),with statistical significance.Compared with the normal group,model group wassignificantly lower in preference,with statistical significance.3Open-field test and to capture reaction:compared with the normal group,modelgroup,open field test score was significantly reduced (P<0.05);the treatment group comparedwith the model group,the score was significantly reduced (P<0.05),with statisticalsignificance;Tianbing Tiaodu capsule in high dose group compared with the modelgroup,there are obvious difference (P <0.05).To capture reaction:model group than in thenormal control group were significantly increased (P<0.05);Tianbing Tiaodu high dose groupthan in model group were significantly reduced (P<0.05).4Step-down test:model group compared with normal group,the latency and errorfrequency difference were different significantly (P<0.05), which was statisticallysignificant;Tianbing Tiaodu capsule in high dose group compared with the model group,therewere significant differences (P<0.05).5Morris water-maze:compared with the normal group,model group rats escape latencyand the original platform number were significant difference (P<0.05); wherein TianbingTiaodu capsule in high dose group compared with the model group,there were significantdifferences (P <0.05),with statistical significance. 6.SS content:after the treatment,compared with model group, the other group SS contentswere significantly decreased (P<0.05),which was statistically significant;Tianbing Tiaoducapsule in the high dose group and the other treatment groups had significant difference(P<0.05),with statistical significance.Conclusion:1The experimental use of "tail clamping angered",through the dynamic observation ofrat the amount of water,sugar preference food intake and emotional behaviors and spatialmemory performance,can better simulate the human liver stagnation syndromemanifestations.2The model of liver-qi stagnation syndrome in rat plasma,hypothalamus and gastricantrum of somatostatin (SS) content increased significantly.3Turbidity toxin method detoxication method, can improve the performance ofstagnation of liver qi stagnation of liver curative effect.4Tianbing Tiaodu capsule can significantly improve the amount of drinking water andfood intake in rats,significantly improve rats emotional behaviors and spatial memorycapacity,and can decrease the plasma of rats, gastric antrum and SS content inhypothalamus,which may be Tianbing Tiaodu capsule used by governor for detoxication intreatment of syndrome of stagnation of neuroendocrine substance one of the basic. |
