Objective Mechanism of plasma cell mastitis (PCM) is still not clear up to now. Thepathological diagnosis is gold standard.This study is designed to find the method ofestablishment a reliable plasma cell mastitis mouse model. It will provide an ideal model forstudy the pathogenesis of plasma cell mastitis.Methods By using the diseased tissue was cut in the surgery of patients with plasma cellmastitis. Mice purchased from Beijing Vitalriver Co. The water-in-oil emulsion is prepared withcomplete Freund’s adjuvant and homogenate which is made from the diseased tissue,it isinoculated to the mouse mammary gland.The sexually mature female BALB/c mice were randomlydivided into5groups: group A (blank control), group B (saline control group), groupC(complete Freund’s adjuvant group), group D(low-dose emulsion group), group E (high-doseemulsion group). Observed for induced situation in3days,7days and14days. The diseased tissue ofmice will be done pathological diagnosis. Statistical treatment of experimental data using statisticalpackages SPSS18.0using chi-square test. In order to test the level of α=0.05.Result In3days there are not successfully established mouse model in group A,group B andgroup C. Group D, there were2mouse model have established. Group E, there were3mousemodel have established. there was no significant difference among five groups (P>0.05). therewas no significant difference of establishing mouse model between group D and group E. In7days there are not successfully established mouse model in group A,group B and group C.Group D, there were4mouse model have established. Group E, there were7mouse modelhave established. there was significant difference among five groups (P<0.01). there was nosignificant difference of establishing mouse model between group D and group E (P>0.05). In14days not successfully established mouse model in group A,group B and group C. In groupD, there were4mouse model have established. In group E, there were9mouse model have established. there was significant difference among five groups (P<0.01). there was nosignificant difference of establishing mouse model between group D and group E (P<0.05).Pairwise comparisons with inoculation of the length of time,in group D between in3days andin7days or in14days,and between in3days and14days, there was no significant differenceof establishing mouse model(P>0.05);in group E between in3days and in7days, there wasno significant difference of establishing mouse model(P>0.05); in group E between in3daysand in14days, there was significant difference of establishing mouse model(P<0.01); ingroup E between in7days and in14days, there was no significant difference of establishingmouse model(P>0.05).Conclusion1. In this study, human plasma cell mastitis-like pathological changes can beinduced in female BALB/c mouse mammary gland which inoculated with human plasma cellmastitis tissue homogenates and complete Freund’s adjuvant suspension.2. The PCM mousemodel which was established by inoculation with emulsion,were positively correlated with thedose and time.
