Objective:The purpose of this study is to detect the localization of aquaporin9(AQP9) proteinand expression of AQP9mRNA and its protein in human placenta and fetalmembranes of patients with preeclampsia(PE),discuss the role of AQP9in thepathogenesy of preeclampsism.Methods:A total of40womem with preeclampsia and40normal pregnant womem as controlparticipated in this study.All were admitted to Fujian Maternity and Children HealthHospital for delivery from April2010to May2011. Patients with PE were dividedinto mild(n=20) and severe (n=20) group. All patients gestational weeks were37to41weeks. Immunohistochemical EliVision determined expression positioning ofaquaporin9(AQP9) in the placenta and fetal membranes. Reverse transcription-polymerase chain reaction (RT-PCR) detected mRNA levels from mild and severepreeclampsia patients (case group) and normal full-term pregnancy (control group)placenta and fetal membranes in aquaporin9(AQP9) expression; Semi-quantitativemeasure of AQP9by Western-bloting, taking β-actin as the control protein,and thegray scales are quantitated by Quantity One4.6.2to measured expression levels ofAQP9in all patients with placenta and fetal membranes.Results:1. AQP9in the two groups of patients expressed in the placental syncytiotrophobl-ast, chorionic trophoblast cells, amniotic epithelial cells,and the placenta vascularwall in preeclampsia was also expressed.2.Aquaporin9(aquaporin9, AQP9) mRNA in the human placenta and fetal membran es were all expressed. All samples were amplified β-actin75bp bands, AQP9mRNAamplified fragment of76bp.3.Normal and preeclampsia placenta AQP9ratios of gray scale were0.19±0.01,0.53±0.13, fetal membranes were0.16±0.01,0.53±0.15, Two groups AQP9ratios ofgray scale were0.41±0.01,0.66±0.02, placenta and fetal membranes of twogroups were statistically significant compared to ratios of gray scale (P <0.01); Mildand severe preeclampsia placenta ratios of gray scale were0.38±0.02,0.67±0.01,two groups of the placenta, fetal membranes were statistically significant compared tothe ratios of gray scale (P<0.01).Conclusion:1.AQP9was mainly expressed in the placenta syncytiotrophoblast, amniotic epithelialcells, chorionic trophoblast cells, and the placenta vascular wall in preeclampsia wasalso expressed, suggesting that AQP9is involved in the energy metabolism ofPE,and then participates the occurrence of preeclampsia.2. AQP9mRNA in the human placenta and fetal membranes were expressed. AQP9protain in the preeclampsia placenta and fetal membranes was higher than normal patients,and severe preeclampsia patients than mild preeclampsia patients, this showsthat increased expression level of AQP9in human placenta and fetal membranes maybe involved in the pathogenesis of preeclampsia.
