| Macleaya cordata (Willd) R. Br. is a species from genus Papaveracea e, Macleaya plants. The main chemical component parts of Macleaya is al kaloid. It has some pharmacological actions, such as inhibition, disinsection, antitumor. The purpose of this research is isolation and structure elucidati onof Macleaya leaves, and discuss the reaction process and reaction produc t of protopine alkaloid that is the highest content alkaloid and acid. The re sults were obtained as follow:1.The chemical composition pretesting of Macleaya leaves:After testing the chemical composition of Macleaya cordata by system pretesting method, the results showed that there were alkaloids, phenols, sterides, flavone and glycosides, organic acid, oil, coumarin lactone and glycosides, cardiac glyc oside and protein.2. Extraction and separation of chemical ingredient:Macleaya leaves ar e extracted by ethonal after cold-leached extract with petroleum ether. Then three polar parts are prepared by means of solvent extraction, which respec tively are the part of chloroform(W3), the part of n-BuOH(W4) and the par t of water(W5). The part of chloroform(W3) are isolated by TLC Analysis as further separate parts. Nine compounds were identified with column chr omatography and re-crystallization.3. Structural determinations of 9 compounds:9 compounds are detemine d as derivatives of 6-acetonyl-dihyrosanguinnarinc(1), Dihydrosanguinarine (2), Dihydrochelerythrine(3), Sanguinarine(4), Oxysanguinarine(5), Chelerythr ine(6),5-O-feruloyquinic acid methylester(7), Protopine(8), Allocryptopine(9). Compound 7 is a new natural product.4. Structural discussion of the main composition of Macleaya leaves for ming salt with acids:Protopine alkaloid form salt with acids, such as ben zoic acid and salicylic acid. The structures of the salts were identified by means of 13C NMR, 1H NMR,’H-’H COSY, HSQC, HMBC. |
