Objective:The active ingredients of Chinese herbal medicine are usually distributed in between the cytoplasm of the cell and cells, and most of them are in cells. If the cell wall is broken, which is beneficial to dissolution of active ingredients of crude drugs.Ultrafine is the final product of ultrafine grinding, and the breakage rate is up to 90%.Hence ultrafine have some special physical and chemical properties, such as solubility, dispersion, adsorption, chemical reactivity, etc. Superfine powder technology is introduced to Chinese medicine field, which produced micron medicine. However, the application of superfine powder in Chinese pharmaceutical industry is still at the exploratory stage. We should perfect the powder quality standardization system, and strengthen the systematic study of the powder preparation process technology, quality standards and the major pharmacodynamics. In this thesis, as the research object of Isatidis Radix, author explore preparation of Isatidis Radix, the fingerprint of multi-component and the antiviral activity of different polarity parts, which provide experimental data for formulating and perfecting the quality standards of Chinese medicine ultrafine.Methods:(1) The study on preparation technology of Isatidis Radix ultra-fine powder:the ultra-fine timeã€temperatureã€water contentã€medium fill rate index were selected by using D50 value measured. (2)The study on characterization and physical properties of Isatidis Radix ultra-fine powder:The power is characterized by appearance, microstructure, powder bulk density and other aspects; The changes to liquidity and moisture absorption of different powder particle size are determined by angle of repose, the moisture rate, moisture percentage, CRH and so on; The chemical compositions of different powder particle size are compared by IR.(3) The study on fingerprint of ultrafine: Common model fingerprint were established as a index of adenosine, epigoitrin and 15 kinds of amino acids.(4) The study on antiviral activity of powder:Six effective parts of Isatidis Radix are obtained by using the system solvent extraction, and the effective anti-viral activity of every part is compared.Results:(1) The optimization ultra-fine condition of Isatidis Radix was determined:crushing time was 16min,temperature was-10~10℃,water content was 6~8%,medium filling rate was 60~70%;the optimal particle size distribution was D50≤13μm.(2) The results of physical properties show that:When Isatidis Radix is crushed into ultra-fine powder, appearance color trend to light, texture trend to more uniform, mobility reduce and moisture increase;Microscopic identification showed that organelles were broken and common features of microscopic identification missed;IR spectra of different particle size powders were the same. (3) The HPLC fingerprint of water-soluble components in ultrafine of 13 different areas was established, and 7 common peaks were confirmed by using the gradient elution, similarity of 13 batches of samples were more than 0.70;The HPLC fingerprint of amino acids components in ultrafine of 13 different areas was established, and 15 common peaks were confirmed by using the gradient elution, similarity of 13 batches of samples were more than 0.70.(4) Antiviral activity in vitro showed that, antiviral activity of parts of chloroform was the strongest;the concentration of completely inhibiting virus replication of Isatidis Radix ultrafine was significantly less than the powder, and it was evident that the bioavailability of superfine powder greatly increased, thereby which provided experimental basis for the saving medicine.Conclusion:With the ultrafine powder and ordinary powder of Isatidis Radix comparison, the appearance, the texture, the aerated density, the fluidity, the humidity absorption have greatly changed, however, major chemical compositions have not obviously varied. From the anti-virus test, we knew, the ultra-fine process of Isatidis Radix can improve the bioavailability and efficacy, which verify the superiority of ultrafine powder and it play an important role in effects of TCM. |
