The objective of this study was to research the expression of tumor protein p53(Tp53) in yak(Bosgrunneins) during different development period of embryo and to verify the relationship with in vitro fertilization(IVF) embryo development. Gather the ovarian of female yak, Collect yak class A and class B oocyte during in vitromaturation and to culture these IVF embryos. The specific primers were on the basis of conservative Tp53 gene sequence of cattle,.The expression of Tp53 in different periods of IVF embryos and Ovary were detected by the methods of PCR, quantitative Real- time PCR and immunofluorescence at m RNA and protein levels. analyzed its gene structeue and bioinformatics. Research indicates:(1)Tp53 mainly present in the nucleus and small amounts in the cytoplasm, its role and DNA damage.(2)This is the first successfully cloned the full length of 1287 bp yak Tp53 gene c DNA, and to submit the gene sequence to Gen Bank, KR185965.(3)Homology analysis showed that the corresponding gene sequence of yak and zebu Tp53 highest homology, indicating that Tp53 gene is highly conserved in evolution.; Yak Tp53 protein homology modeling the formation of a spiral pleated sheet surrounded by α β 3D structures.(4)The expression of Tp53 genes at different times of ovarian is related.to granulosa cells and the conversion of apoptosis and ovarian endocrine.From this study, Tp53 gene was expressed in the early embryo and yak different rut periods ovaries, indicating Tp53 gene plays an important role in the reproductive process.The relationship between the expression of Tp53 and developmental block is positive correlation,, its role is related to DNA damage. why the expression of Tp53 is different may be due to the extent of DNA damage and endocrine factors in different estrus period ovaries. The successful cloning and bioinformatics analysis in Yak Tp53 lay a foundation for the further study.
