With the further studies on embryo engineering technique,commercialization and application, the technique of Ovum Pick Up and in vitro fertilization have become efficient ways to solve the problems of embryo shortage . but the present study have not achieved the aim on efficient production.The study was to establish the droplet culture method to satisfy the feather of Ovum Pick Up technique such as obtain fewer and lower qualitity oocytes in order to improve the efficience of in vitro production embryos the study optimize the system of in vitro culture from five aspcet as following different states of ovaries, different semen, fertilization time, season and co-culture for in vitro culture of bovine with droplet methodThe paper were conducted to study the effects of medium drop size on developmental competence of bovine oocytes derived from slaughterhouse ovaries in vitro maturation,fertilization,culture fertilization droplet.and culture medium for IVF covered with paraffin oil and mineral oil.The results showed that different drop size(70ul,150ul) had no significant diffeneces in cleavage rate (67.2%, 64.2%)and blastocyte rate(25.4%, 24.3% )after IVF. Drop size (70ul)covered with paraffin oil or without had significant difference in cleavage rate and had (59.3%, 70.3%) little difference in blastocyte rate(27.1 %, 21.4%) the result was advantageous to control group and better than the control group in early embryo covered with paraffin oil on culture medium of 200μl.the cleavage rate (72.3%,70.5%) and blastocyte rate (33.2%,22.3%) are significant different and better than control group so in vitro culture of few oocytes . we advice to use droplet culture,in a word it is received better culture effect covered with paraffin oil than mineral oil.The paper also discussed'the effects of oocytes derived from slaughterhouse different states of ovaries, different semen, fertilization time, season and co-culture for in vitro culture of bovine with droplet method. The result indicated that (1) After culture the oocytes derived from slaughterhouse different states of ovaries, there were no significant difference in cleavage rate and blastocyte rate (P>0. 05) .(2) There were significant difference in cleavage rate and blastocyte rate in different semen(P<0. 05) (3) The effect on fertilization time of 12h is better. It had significant difference compare to 6h,8h and 16h(P<0. 05).(4) Spring and autumn had better effect than summer and winter on in vitro fertilization(P<0. 05). (5) Granular cell layer can improve the blastocyte rate obviously, but can not improve the cleavage rate (P<0. 05).
