Potato(Solanum tuberosum L.) is known as an important crop-with existing many wild diploid species. Self incompatibility is an important limiting factor for the utilization of diploid wild species. Using three inbred populations of diploid potato originated from Solanum chacoense as materials, named 11H-129-13,11H-129-34 and 11H-129-51, which have been self-pollinated to the 8th generation, the study focused on probing the genetic mechanism of self-compatibility, morphological observation, cytoplasmic background detection and SNPs based test of homozygosity of the populations. The main results were as follows:(1) The ratios between self-compatibility and self-incompatibility were 3:1 in population 11H-129-13, 2:1 in population 11H-129-34, and 2:1 in population 11H-129-51. Thus, we could preliminarily speculate that: the self-compatibility and self-incompatibility traits in these inbred populations might be controlled by multiple genes under a single locus. The results also showed that the averaged number of seeds contained in each fruit was only 37 TPSs, with lower germination rate of 36%,seedling rate of 4.5% and flowering rate of 55%. The typic characteristic of the three inbred populations showed stunt but no other obvious separation in morphology.(2) The cytoplasmic type of all selfing inbred lines is W/α, which is identical to the maternal parent, but different from the dominated cytoplasmic types(W/α [D]and T/?) of cultivated tetraploid potatoes in Yunnan province.(3) The results of SNPs analysis of the whole gemonic resequencing data revealed that the degree of homozygosity in the three selfing inbred populations showed obvious difference from each population and difference from each chromosome of haploid genome at the 8th selfing pollinated generation.The above preliminary results provided the basis for the establishment of inbred population of diploid potato and it’s further utilization in breeding studies.
