Casuarina trees have resistance to the typhoon, tsunami, waves erosion prevention, sand-fixation, alkali, or poor features, was to restore the southern coastal area of choice for desertification causes the degeneration of ecosystem.A hydroponic cuttings was one of the main means of reproduction casuarinas. But because of difference of hydroponic rooting rate between different tree species, limited the development and utilization of choiceness clones and individual trees. In this study, appropriate hormone and optimal concentration for water-cultured rooting of Casuarina glauca, C.cuninghamiana, C.junghuniana and rooting ability of various provenances were tested. Meanwhile, the rooting mechanism of Casuarina trees were revealed by monitoring of the content of nutrients and oxidation enzymes activity. In order to choose the rooting ability of casuarina excellent provenance and asexual propagation, utilization to provide theoretical reference.The results were as follows:(1) The hormone of ABT-1 in concentration of 100 mg·L-1 was the best treatment in rooting ability of C.glauca,C. cuninghamiana and C.junghuniana, their rooting rate of 90%, 96.67% and 100% respectively.(2)Under the condition of best water-culture rooting, There were very significant differences in rooting ability traits among C.glauca、C. cuninghamiana and C. junghuniana provenances, genetic coefficient variation change in the range of 11.33% to 26.06%,15.66% to 29.12% and 15.69% to 32.11%.(3)Cglauca> C. cuninghamiana and C. junghuniana provenances material had the high broad heritability, rooting rate, rooting number, The longest root length, rate on deviated lateral root controlled by the strong genetic factor, have the potential for genetic improvement.(4)Based on the method of subordinate function.Seven provenances of C.glauca were ranked in rooting ability according to subordinate function values as follows:13128> 13146 > 13142> 13144> 13139> 13143> 13141, the rooting rate ranges were from70% to 100%.Eleven provenances of C.cuninghamiana were ranked in rooting ability according to subordinate function values as follows:13125>13129>13511>13514>13519>13518 >14005>13127>13156>13508>13149, the rooting rate ranges were from 20.0% to 96.67%. Six provenances of C.junghuniana were ranked in rooting ability according to subordinate function values as follows:18951> 18948> 18950> 18954> 18952> 18949, the rooting rate ranges were from 76.67% to 100%.(5)Soluble sugar content and soluble proteincontent of casuarinas in the process of determination results indicate that:the content of nutrients as the water-culture time change regularity. Among them, soluble sugar content of 4 kinds of casuarinas and soluble protein contentof C. equisetifolia,C. cuninghamiana cuttings had same trend. In adventitious roots induction period, levels drop; in adventitious root formation, content were rised, with adventitious roots induction period,soluble protein content of C. glauca and C. junghuniana downed before they rise; in the wake of a massive formation of adventitious roots, soluble protein content was still falling.Adventitious roots induction period, after ABT1 treatment, soluble sugar contentof 4 kinds of casuarinas and soluble protein content of C. equisetifolia, C. cuninghamiana decreased than control. In the wake of a massive formation,after ABT1 treatment of 4 kinds of casuarinas,the change trend of soluble sugar content in greater than the control, after ABT1 treatment,soluble protein content of C. equisetifolia,C. cuninghamiana were increased greater than the control. after ABT1 treatment, soluble protein content C. glauca, C. junghuniana was always higher than the control.(6) Casuarinas water-culture physiological activity in the process of the index determination showed that:peroxidase (POD), polyphenol oxidase (PPO) and the activity of indole acetic acid oxidase (IAAO) with water-culture time change regularity. Adventitious roots induction period, POD, PPO activity was rised of four kinds of casuarinas; Adventitious root formation, POD activity first rise after falling of 4 kinds of casuarinas. PPO activityof C. equisetifolia, C. cuninghamiana downed after rised first, PPO activity of C. glauca and C. junghuniana had down before they had rise; in adventitious roots growth period, POD and PPO activity of four kinds of casuarinas declined. In adventitious roots induction period, IAAO activity of C. equisetifolia, C. cuninghamianaand C. junghuniana had rise, IAAO activity of C. glaucahad declined. In adventitious root formation period, IAAO activity of C. equisetifolia, C. cuninghamiana, and C. junghuniana began to decline, IAAO activity of C. glauca increased.ABT1 processing of 4 kinds of casuarinas, POD, PPO activity were always higher than the control. ABT1 processing of C. equisetifolia, IAAO activity has been higher than the contrast; ABT1 processing of C. glauca and C. cuninghamiana, IAAO activity than controls; ABT1 processing of C. junghuniana, IAAO activity basically samed with contrast.
