A Preliminary Study On The Traditional Chinese Medicine And Western Medicine Treatment Of Mycoplasma Bovis Disease

In recent years, Mycoplasma bovis (M.bovis) disease presented trends in our country and Mycoplasma bovis disease also showed an upward trend in Ningxia. In this study, the drug sensitivity test in vitro were tested on the typical M.bovis strain and the new isolate strain, with the antibiotic and herb extract. Based on the drug susceptibility results, the compatibility of medicines and herb extract, effect on M.bovis, were selected respectively, and the curative effect was evaluated on the rabit infection model. At the same time the safety of the compatibility of herb extract was evaluated on the mouse. The study includes:1. To evaluate the curative effect of antibiotics, two M.bovis strains were tested for in vitro drug susceptibility, and the results were anaryzed as:Mycoplasma bovis is sensitive to minocyc line,tetracycline,spectinomyc in,Acetylspiramyc in,tiamulin,enrofloxacin,c iproffox acin, norfloxacin, kanamycin, novobiocin, neomycin; moderately sensitive to tylosin McGrady vancomycin, levofloxacin, Tai Wan streptozotocin; resistant to lincomycin, azithromycin, erythromycin, clindamycin, clarithromycin, ofloxacin, polymyxin B. Further joint drug susceptibility experiments were tested to the typical strain from Ningxia, which are extremely sensitive to M.bovis, and 3 formulas of antibiotics were got. Formula A includes tetracycline hydrochloride;formula B includes:spectinomycin hydrochloride;formula C includes:Ling cough lung (30% tiamulin), the pharmaceutical compositions of A+B, A+C, A+B+C. The study on formula of antibiotics provided theoretical and practical knowledge to the clinical treatment for M.bovis disease.2. In order to solve the problems of resistance to antibiotics, drug residues, etc., noval herbal medicine for M.bovis is required, so in this study the drug susceptibility experiments with herb extract were done, and the results showed that M.bovis was extremely sensitive to berberine; moderately sensitive to Houttuynia, cloves, Speranskia; sensitive to Cork, rhubarb, cow heart komarovii, licorice, picrorhiza, Sophora, Cnidium, skullcap, turmeric, pseudolaric, pepper; resistant to ground child skin, Radix, Andrographis. Accourdig to the knowledge of Chinese traditional medicine and the drug sensitivity results, a noval formula of herbs were made, and it includes:Sophora, Houttuynia, rhubarb, Treats, Speranskia, Hu berberine, pepper, berberine, etc., then the compound herb with the formula was extracted, and the solution was lr/ml,the compound herb will be used for the treatment forM.boivis disease.3. The rabit infection model was built in this study, to improve the effectivity of curative effect evaluation experiments. The clinical symptoms, pathological autopsy tissue sections and display cases showed that the rabits were infected M.bovis via intranasal approach. With the rabit infection model, the curative effect experiments should be started at the 7 day after infection.4. To fiding the best therapy plan for M.bovis disease,3 groups were set, with formulated biotics, compound herb and combination of both formulated biotics and compound herb repectively. The curative experiments showed the results:the tetracycline hydrochloride in 0.1 g per kg weight+ spectinomycin hydrochloride in 0.1 g per kg weight+ kechuanfeiling in 0.1 g per kg weight group had the best effect in all the antibiotics treatment group. Homemade Chinese medicine prescription nasal spray is ideal for M.bovis disease treatment effect, and choose to medicine dose of 100 mg per kg weight to Chinese medicine dose. Effect of combination of Chinese and western medicine treatment group is higher than the noval herbal medicine treatment group, but no significant difference.5. In order to evaluate the applied reliability and ascertain the toxicity of the traditional Chinese herbal compound medicine nasal spray, the experiment of mucosal irritative test and acute toxicity were did.The results of mucosa stimulating test were negative and the determination of LD50 of the traditional Chinese herbal compound medicine nasal spray was conducted in mice with different doses.The result showed that the LD50 of traditional Chinese herbal compound medicine nasal spray was 41438mg per kg weight, and the 95 percent confidence interval was from 39490.26 to 43482.05 mg per kg weight, the standard error Slog LD50 was 0.01067. According to the toxicological evaluation standard, the medicine belongs to the non-toxic matter,the results confirmed that the self-made traditional Chinese herbal compound nasal spray was safe and non-toxic,and it can use security in clinical practice.
