Mycoplasma bovis is an important cause of pathogenic microorganisms.In the recent years,M.bovis respiratory disease is causing huge economic losses to the aquaculture industry at local and abroad,but the research on M.bovis immunity at local and abroad is not enough,and there is no specific medicine to cure.Immunization is one of the important way of prevention and control of M.bovis disease.In this study we successfully prepared the M.bovis inactivated vaccine,based on evaluate the immune effect of rabbits on the basis of the experimental calves were immune effect evaluation system,carried out the evaluation of immune effect of inactivated M.bovis vaccine immune calves after the prevention the effect is obvious.The main research contents are: 1.Serological detection of Mycoplasma bovis in Yongning County of the Ningxia Hui Autonomous Region.10 large-scale cattle farms in Yongning County were randomly selected for the detection of Mycoplasma in different months according to the data of 312 samples.The results showed that the mycoplasma bovine positive number is 33.65%(105/312).The highest positive rate of Mycoplasma was 100%,the lowest positive rate was 7.41 A group of positive rate was 100%;the positive rate of Mycoplasma positive rate is much higher than that of the dairy cattle market,the positive rate of Mycoplasma bovis West F1 hybridization was the lowest,the difference between the West and Holstein crossbred cattle F1 positive rate of Mycoplasma bovis significantly(P<0.05)Angus cattle and cattle species Qinchuan were not significant(P> 0.05);Mycoplasma bovis disease mostly prevalent in April.The above data showed that Mycoplasma in Yongning County have different levels of popularity,other cattle strong resistance ability of West F1 hybrid cattle bovine mycoplasma relative test,but it may also with the introduction and breeding environment.2.Extraction and screening of traditional Chinese medicine and animal toxicity testAccording to the disease of Mycoplasma bovis and the therapeutic effect of traditional Chinese Medicine,drug sensitivity test in vitro proliferation of Oxford cup method in vitro drug sensitivity test and drug sensitivity agar 21 herbs combined with primary incompatibility with the addition and subtraction principle into prescription of traditional Chinese medicine contains 12 herbs Coptis,rehmannia,clove etc.prescription.The mucous membrane irritation test and acute toxicity test to evaluate the safety of self-made compound Chinese medicine preparation,mucosal irritation test showed that the compound preparation of no stimulation of the mucosa,the drug intraperitoneal injection of the acute toxicity tests of half lethal dose of confidence limit range is 39490.26 ~ 43482.05mg/kg,standard error Slog LD50=0.01067,far less than the toxicity of drug toxicity the national standard,confirmed that the drug is safe and non-toxic,so safe in clinical use.3.Preparation of traditional Chinese medicine and evaluation of therapeutic effect of M.bovis.Mycoplasma bovis mostly cause respiratory diseases in the calf,antibiotics are commonly used drugs,and now a days there is also drug resistance against antibiotics,Calves suffering with Mycoplasma bovis,eye disease symptoms,diagnosis and cure is very difficult.Therefore,in this study,Rhizoma Coptidis,Radix Sophorae Flavescentis,Houttuynia,rhubarb,phellodendron,et al.used 12 kinds of traditional Chinese medicine preparation,antibiotic treatment test on calf infected by Mycoplasma bovis.The laboratory isolates of NP2 strain of Mycoplasma bovis infection in animal experiments successfully,making animal model test to observe the clinical symptoms,physiological indexes,necropsy and histological examination,the results of the study show: after to the negative control group outcome of traditional Chinese medicine in the treatment of infection of Mycoplasma bovis calf viscera and tissue,i.e.to gradually eliminate the symptoms and rehabilitation;antibiotic treatment of calf organs continue to suffer erosion of Mycoplasma bovis,showed clinical symptoms of the positive group.The results showed that the traditional Chinese medicine preparation has significant effect on the treatment of Mycoplasma bovis,and no drug resistance.4.Preparation of M.bovis inactivated vaccine and evaluation of immune effect in cattleIn order to study Mycoplasma bovis formalin inactivated strains with white oil adjuvant through the preventive effect of intramuscular injection of Mycoplasma bovis disease,the establishment of PC group,NC group and inactivated vaccine clinical trials.Through the clinical symptoms,ELISA technology for specific antibody level detection,pathological examination and histopathological analysis of the vaccine immune effect evaluation.The results showed that the inactivated vaccine group and normal body temperature,NC group showed no significant difference(P>0.05),the antibody level in first to two weeks after the last immunization was visible pathological changes of pathological examination and no gross analysis,lung and tracheal tissue and normal tissues.The results showed that the inactivated Mycoplasma bovis vaccine has a good effect on the prevention of Mycoplasma bovis disease,and it has a great reference value for the study of the vaccine.
