Studies On Monosaccharide Composition Analysis Of Polysaccharides In Ziziphus Jujuba Mill Cv. Lingwuchangzao Fruit

In this experiment, Ziziphus jujuba Mill cv. lingwuchangzao fruits as experimental material, using the methods of hot water extraction, spectrophotometry and gas chromatography-mass spectrometry to research polysaccharides extraction and composition analysis of Ziziphus jujuba Mill cv. lingwuchangzao fruits in different developmental periods and different types respectively, aiming to explore the change rule of its polysaccharides content and monosaccharide composition. The results showed that:1. Crude polysaccharides in Ziziphus jujuba Mill cv. lingwu-changzao fruits of different developmental periods and different types were extracted by hot water extraction, obtaining ratio of the early bulking period, the rapid enlargement period, the fruit coloring period and the fruit maturation period was 0.778%,0.865%,0.983% and 1.795%, respectively. Indicating that crude polysaccharide yield presented ascendant trend with the development of the fruits. Obtaining ratio of the ordinary type, the big-fruit type, the early maturity type and the inermous type was 1.795%, 1.504%,0.999% and 0.956%, respectively.2. Contents of crude polysaccharides were detected by phenol-sulfuric acid method, the results showed that the contents of crude polysaccharides showed a trend of rising with the development of fruits, reaching the peak in the fruit maturation period, at 59.77%. The order of crude polysaccharides content of different types from high to low was ordinary type, early maturity type, big-fruit type and inermous type.3. Four kinds of polysaccharide like as Ju-0, Ju-1, Ju-2 and Ju-3 were all obtained from Ziziphus jujuba Mill cv. lingwuchangzao fruits both in different developmental periods and different types by DEAE-52 cellulose ion-exchange columns, all of which were estimated preliminary to be homogeneous compositions by using TOYOPEARL HW-55S Molecular size exclusion chromatography purification and ultraviolet scanning detection.4. The refined polysaccharide content of Ziziphus jujuba Mill cv.lingwuchangzao in different developmental periods and types had a large difference because of component difference. As was shown below, the Ju-0 content of Ziziphus jujuba Mill cv. lingwuchangzao fruits in different developmental periods from higth to low was the rapid enlargement period, the fruit coloring period, the fruit maturation period and the early bulking period. The Ju-1 content was:the fruit coloring period, the rapid enlargement period, the same as the fruit maturation period and the early bulking period. The Ju-2 content was:the early bulking period, the fruit maturation period, the rapid enlargement period and the fruit coloring period. The Ju-3 content was:the fruit maturation period, the early bulking period, the rapid enlargement period and the fruit coloring period. With regard to Ziziphus jujuba Mill cv. lingwuchangzao fruits of different types, the ordinary type had the highest content of the Ju-0 and Ju-3, the big-fruit type had the highest content of the Ju-l and the early maturity type had the highest content of the Ju-2.5. The GC-MS analysis showed that the contents of Arabinose, galactose and rhamnose were higher, followed by glucose, mannose and xylose in the different developmental periods and types ofZiziphus jujuba Mill cv. lingwuchangzao fruits. In addition, we known that the refined polysaccharide in Ziziphus jujuba Mill cv. lingwuchangzao fruits also contained ribose, fucose, glucuronic acid, galacturonic acid and did not contain fructose at all by compared with NIST mass spectrometry library.
