In order to overcome decreasing forest planted land of ginseng and other shortcomings of conventional breeding a long time, the method of using tissue culture to develop new ways of asexual reproduction ginseng, cultivating new seed varieties may be a new way of ginseng varieties breeding. Ginseng roots, stems, leaves, buds as explants, discussing the optimal disinfection methods and the optimal explant callus induction; In five years, four years, three years and two-year old ginseng stems as explants, discussed the optimal age of ginseng stems explants on stem callus induction of ginseng; In the MS medium alone adding 2,4-D and 2,4-D used with 6-BA influence on ginseng stem callus induction; Ginseng stem callus subculture medium screening; 2,4-D used with 6-BA effect on the adventitious bud induction; Ginseng plantlets proliferation medium screening; The basic culture medium, activated carbon content, the addition of IB A and NAA on the effect of ginseng regeneration seedling adventitious roots induction. The results show that:1.Ginseng root explants disinfection best treatment is 0.1% HgCl2 8 min, the survival rate of 50%; Ginseng buds explants disinfection best treatment is 2%NaC102 24 min, the survival rate of 90%; Ginseng stem explants disinfection best treatment is 0.1% HgCl2 4 min, the survival rate of 70%; Ginseng leave explants disinfection best treatment is 2%NaClO210 min, the survival rate of 45%.2. Ginseng roots, stems, leaves and buds four kinds of explant highest callus induction rate was stems, of 55.71%. In five years, four years, three years and two years of ginseng stem explants, five years ginseng stem callus induction rate was highest, of 54.43%.3. Adding 2,4-D alone can induce ginseng stem callus, MS+2,4-D2 mg/L ginseng stem callus highest induction rate was 56.70%.4.2,4-D and 6-BA, according to ginseng stem callus induction rate to establish the optimal regression equation, when the concentration of 2,4-D was 2.57 (mg/L),6-BA was 0.00 (mg/L). the highest rate of induced 64.75 (%). That 2,4-D was the main hormone, ginseng callus induction and the effect of 6-BA little or almost no effect.5.2.4-D and 6-BA used subculturing ginseng stem callus,10 kinds of medium subcultured the best medium for S7. namely MS+2,4-D 2.00 mg/L+6-BA 0.10 mg/L.6.2,4-D concentration is 2.00 mg/L,6-BA concentration range in 0.00-0.10 mg/L, is conducive to ginseng stem callus induction of adventitious bud. The best ginseng stem callus differentiation medium for MS+2,4-D 2.000 mg/L+6-BA 0.075 mg/L, differentiation rate was 24.44%.7. Generated from ginseng stem callus ginseng adventitious buds proliferation largest number of media was MS+TDZ3.0 mg/L+IBA0.3 mg/L+GA3.0 mg/L, each an average of callus with bud points proliferation 8.1 ginseng plantlets, the fresh weight growth peak 491.7 mg.8. The better basic medium for ginseng plantlets adventitious rooting is 1/2 MS medium andl/2 SH medium; 1/2 MS+IBA2 mg/L+0.1% activated carbon and 1/2 SH+NAA0.5 mg/L+0.1% activated carbon medium adventitious roots induction rate was highest,40%.
