Study On The Establishment Of Ginseng Tissue Culture System

In order to establish a complete ginseng tissue culture system,ginseng sterile leaves and anthers were used as explants,and MS was used as the basic medium.By adjusting 2Z 4-dichlorophenoxyacetic acid(2,4-D),6-furfurylaminopurine(KT)and naphthaleneacetic acid(NAA)concentration of three plant growth regulators to induce leaf callus;adopt uniform design method by adjusting 2,4-D,KT,NAA And 6-benzylaminoadenine(6-BA)in different combinations of plant growth regulators to induce anther callus.The callus of ginseng leaves and anthers were used as explants,and MS was used as a basic medium.By regulating indobutyric acid(IBA),6-BA,NAA,and gibberellic acid(GA3),the plant was grown.Regulator concentration to induce adventitious shoots of leaves and anthers.Adventitious buds of ginseng leaves and anthers were used as explants,and MS was used as the culture medium(excluding ammonium nitrate and potassium nitrate).The uniform design method was used to adjust NO3-,NH4+ and NAA,KT,IBA,6-BA,in Indoacetic acid(IAA)and GA3 were used to induce adventitious roots of different combinations of plant growth regulators.BS was used as a basic medium to induce adventitious adventitious roots by adjusting the concentration of NAA.The main findings are as follows:(1)When using ginseng leaves as explants,the best medium for inducing callus is MS+2,4-D 2 mg·L-1+KT 1 mg·L-1+NAA 0.66 mg·L-1,its induction rate is 96.67%;the best medium for inducing adventitious buds is MS+IBA 3 mg·L-1+6-BA 3 mg·L-1+NAA 3 mg·L-1,its induction rate is 90.00%;the best medium to induce adventitious roots is MS(without ammonium nitrate and potassium nitrate)+NO3-3 mg·L-1+NH4+7 mg · L-1+NAA 0.1 mg·L-1+KT 0.1 mg·L-1+IBA 8 mg·L-1-+6-BA 0.125 mg·L-1+IAA 0.15 mg·L-1+GA3 6 mg·L-1,its induction rate is 95%.(2)When ginseng anthers are used as explants,the best medium for inducing callus is MS+0.56 mg·L-1+2,4-D 0.99 mg·L-1+6-BA 0.99 mg·L-1+KT 1 mg·L-1,and its induction rate is 70%;the best medium for inducing adventitious buds is MS+GA3 40 mg·L-1,its induction rate is 93.33%;the best medium to induce adventitious roots is B5+NAA 2 mg·L-1+8%agar+3%sucrose,the induction rate is 66.67%.
