The application of mice in the study of human and animal biology, physiology, genetics and other aspects is extensive. It is considered an ideal animal model in biomedical areas including cancer, genetic diseases and so on. The study on mass-product ion and protection of transgenic mice becomes more and more important with the increasing interest on it of human. Donor age can affect the quantity and quality of oocytes and embryos produced during superovulation. For the past few years, some imperfection was existed in most study about the effect of mice age on mice fecundity, the groups of weeks and the report of superovulation comparison test of 12-28 weeks mice are very few. Therefore, we groups the mice detailly in this experiment in order to investigate the influence of different ages’mice on superovulation and developmental rate in vitro. The farrowing rate is still not ideal after embryo transfer. The study of transfer embryos and the transfer pattern (single oviduct, double oviduct, single uterus, double uterus) on transfer efficiency still rarely reported. So we made a comparative study on the number of transferred embryos, the stages of transferred embryos and the transfer position. In the selection of mice strains, Kunming mice and C57BL/6 mice were chosen as experimental animal in this research. Because Kunming mice have the advantages of high adaptability and reproductive and survival rate, C57BL/6 mice are international general experimental animals.This paper was divided into two parts. The weeks mice (4-28 weeks of age. a total of 13 groups of four Kunming females; 4-24 weeks of age, a total of 11 groups of four C57BL/6 female mice) were chosen as experimental animals in the part one. All the experiment mice were taken a treatment of superovulation, then we collected and counted the 1-cell and 2-cell embryos, cultured embryos with good quality and observed the developmental situation of fertilized egg in vitro.Kunming mice and C57BL/6 female mice were used as experimental animals in experiment two to discuss the effect of the number of transferred embryos (10,15 and 20), the stages of transferred embryos (1-cell,2-cell and blastula stage) and the transferred position (single oviduct, double oviduct, single uterus, double uterus) on transfer efficiency.The results of experiment one indicate that the number of fertilized egg of collected 8-10 weeks Kunming mice and C57BL/6 female mice were higher significantly than other weeks. The stage of 2-cell embryos development rate in vitro was significantly higher than the stage of 1-cell embryos; Comparing Kunming mice to C57BL/6 mice with different strains but the same week, we found that the difference of the two kinds of mice were significant.The results of experiment two indicate that the number of transferred 20 embryos was higher significantly than 10 and 15 (the group of Kunming mice were 63.75% vs 40.00%, 33.33%; the group of C57BL/6 mice were 62.50% vs 45.00%,46.67%). Average litter size of transferred embryos was significantly difference in 1-cell and 2-cell. Average litter size of transferred 1-cell embryos was higher significantly than 2-cell embryos (the group of Kunming mice were 67.50% vs 48.75%; the group of C57BL/6 mice were 65.00% vs 48.75%). Average litter size of transferred embryos was not significantly difference in 1-cell and blastula. Average litter size of transferred embryos was not significantly difference in 2-cell and blastula. Average litter size of transferred different position was not significantly difference in Kunming and C57BL/6 mice. Average litter size of Kunming and C57BL/6 mice was not significantly difference when transferred same number embryos, same embryo stage and same position.Kunming mice and C57BL/6 mice were chosen as experimental animal in this research. We investigated the influence of different ages’mice on superovulation and developmental rate in vitro. We found ideal age on superovulation. We investigated the effect of the number of transferred embryos, the stages of transferred embryos and the transferred position on transfer efficiency. We found the effective method that could significantly increase birth rate. The research of embryo transfer was established as experiment foundation in future.
