Soybean[Glycine max (L.)] is one of the most important crops, which serves as food, oil, feed and other economic sources. Weed control takes great significance in soybean cultivation. Herbicide-resistant transgenic soybean can help to improve the efficiency and reduce the cost of weed control.In this study, binary vector pCAMBIA1300-35S-g10evo was constructed based on the glyphosate-resistant gene glOevo. Glyphosate-resistant soybean transformants were obtained via Agrobacterium-meditated cotyledonary-node transformation.One of the transgenic lines (GlOevo) was determined to be highly glyphosate tolerant after screening analysis. Western blot detection showed that the 46kDa glyphosate resistant protein was expressed in GlOevo. ELISA analysis showed the EPSPS (5-EnolPyryvyl Shikimic Acid-3-Phosphate Chorismate Synthase) expression level in the second trifoliolate leaf of the V4 stage plants reached 276.73μg/g, which was 0.76% of the total soluble protein. Southern blot analysis indicated that T-DNA was inserted in a single locus in the genome of transgenic line G10evo. Genetic stability analysis showed gene glOevo inherit stably between different generations of G10evo. Glyphosate spraying analysis at V3 stage plants in the green house showed that G10evo was tolerant to a high level of glyphosate (4 times of the recommended field concentration). All these results indicate that G10evo is high resistant to glyphosate, thus being promising for commercial use.
