Piglet yellow dysentery is induced by enterotoxigenic Escherichia coli, with high morbidity and mortality, and caused a great economic loss to the swine industry. At present using antibiotics and chemical drug is the main way to treat piglet yellow dysentery in clinical practice. But these drugs have been used for a long time, making the problems of bacteria resistance and drug residues in meat product become more and more prominent. Now with the progress of science and technology, pollution-free production has become a trend, the prospects of using antibiotics and chemical drugs will be greatly restricted. Traditional Chinese medicine is a promising alternative to antibiotic with the advantages of wide antibacterial spectrum, enhancing the immunity, no residue etc, and accord with the requirement of green pig industry. From the items of toxicology and pharmacodynamics, the clinical efficacy and safety of Yujin San decoction were investigated to provide theoretical basis for the further clinical application.In order to test the safety of Yujin San decoction, the acute toxicity was examined. Preliminary experiments: single dosage 5.0 g/kg can’t cause the death of mice and the median lethal dose(LD50) can not be obtained. Then the maximum concentration and maximum volume of drug were given to the mice in a single test, and there was no death in the mice. Finally we gived mice the maximum concentration of drug many times in a day, and did not find the mice poisoning or death. The results indicates that Yujin San decoction has good safety.In order to further test the long-term safety of Yujin San decoction, we performed the subchronic toxicity. The experimental rats were divided into four groups, respectively, high dose group, middle dose group, low dose group and the control group. Rats in the drug group were given drug once a day for four weeks, and the control group were given the same volume of normal saline.The results showed that Yujin San decoction have no adverse effect on body weight, feed intake, water intake, viscera coefficient, hematology and blood biochemical indexes of rats indicating that Yujin San decoction has good safety.In order to test the therapeutic effect of Yujin San decoction, the model of mice diarrhea was established. Fifty mice were randomly divided into five groups, high dose group, middle dose group, low dose group, model group and normal saline group. The mice in the drug group were given drugs two times one day, and the mice in the control group were given the same volume of normal saline. The results showed that Yujin San decoction improved the body weight and the pathological changes of internal organs in mice of diarrhea, and reduce the amount of inflammatory cells in mice of diarrhea, suggesting that Yujin San decoction has good therapeutic effect on mice diarrhea.In order to test the clinical efficiency of Yujin San decoction, the clinical piglet yellow dysentery cases were treated. First, the clinical symptoms, autopsy, isolation and identification of bacteria were used to select clinical cases of piglet yellow dysentery. The clinical cases were divided into five groups, respectively, high-dose group(2 g/kg), middle dose group(1 g/kg), low dose group(0.5 g/kg), the control group and enrofloxacin group. The results showed that the isolated bacteria were pathogenic Escherichia coli. The efficiency of the high and middle dose group is 95% by giving medicine for five days, similar to that of enrofloxacin group. The cure rate of high dose group is higher than that of enrofloxacin group.In summary, Yujin San decoction has good safety and has obvious therapeutic effect on piglet yellow dysentery by giving medicine for five days. It provides theoretical basis for further clinical application of this Chinese herbal formula.
