Study On Zhizi Baipi Decoction And Huang Qin Zixiening In Treating E.coli Diarrhea In Piglets

Diarrhea is a common disease in the pig breeding industry.It occurs frequently in piglets under 30 days of age,and it will bring huge economic losses to the pig breeding industry every year.At present,the common method for treating piglet diarrhea is still antibiotic treatment.However,due to drug resistance and the problem of antibiotic residues in pork,the use of antibiotics is becoming more and more restricted.Therefore,the application of Chinese medicine in the veterinary field due to its environmental protection and no residue.Therefore,research and development of Chinese medicine preparations that can treat piglet diarrhea are of great significance to the national economic development and food safety.Zhizi Baipi Decoction comes from "Treatise on Febrile Diseases",which is a traditional prescription in China.It is often used to treat jaundice,but its application in treating diarrhea has seldom been reported.In this paper,the minimum antibacterial concentration and orthogonal design were used to optimize the extraction process of Zhizi Baipi Decoction and to determine the antibacterial effect of it.The effect of Zhizi Baipi Decoction on diarrhea in suckling mice was established by establishing a model of diarrhea in suckling mice and administering the drug.Investigate its therapeutic mechanism in immune function by measuring its active ingredients effect on the proliferation and phagocytosis of macrophages.By establishing a piglet diarrhea model and administering the treatment,the clinical application effect of the Chinese medicine monomer ratio formulations(named Huang Qin Zixiening)screened by the research group on piglet diarrhea was studied.In this paper,the orthogonal design method is used to optimize the ethanol reflux extraction process of Zhizi Baipi Decoction by examining the minimum inhibitory concentration of its extract.The best extraction processes were that the extraction temperature was 80℃,the number of extractions was 2,the concentration of ethanol was 60%,the material-liquid ratio was 1:12,the extraction time was 1 h.Under these conditions,the minimum bacteriostatic concentration of Zhizi Baipi Decoction was 6.25mg/m L,and the extraction rate was 25.9%.In vivo experiments to study the therapeutic effect of Zhizi Baipi Decoction extract on ST enterotoxin diarrhea model in suckling mice.The results showed that the optimal dosage of Zhizi Baipi Decoction for the treatment of diarrhea in suckling mice was 2g/kg,while the highdose group with a dosage of 4g/kg was less effective.We speculated that it may be caused by toxic impurities in the extract of Zhizi Baipi Decoction.We used the optimal dosage to treat different degrees of diarrhea,which has obvious therapeutic effects on mild and moderate models,indicating that the extract of Zhizi Baipi Decoction may reduce ST enterotoxin activity or repair the ST enterotoxin-induced metabolic disorders,inhibiting the production of intestinal fluid to treat diarrhea.Our preliminary treatment of piglet diarrhea was performed using Zhizi Baipi Decoction extract.The results show that the administration of its extract can improve the symptoms of piglet diarrhea to a certain extent,and the combination with enrofloxacin can shorten the treatment cycle of enrofloxacin.By studying the effect of the active ingredient monomers of Zhizi Baipi Decoction on the proliferation and phagocytosis of macrophages,the mechanism of immune ingredients of it in the treatment of diarrhea was explored.The results showed that geniposide,crocetin-I and liquiritin could enhance the phagocytic ability of macrophages,indicating that Zhizi Baipi Decoction may regulate the body’s specific and nonspecific immune functions by enhancing macrophage activity to treat piglet diarrhea.We established a piglet E.coli diarrhea model and treated it with different concentration ratio preparations.We used feces as indicators to evaluate the clinical application effect of the Huang Qin Zixiening preparation screened by the research group on piglet diarrhea.The results showed that the Huang Qin Zixiening can effectively treat piglet diarrhea caused by ST enterotoxin,with an effective rate of 100% and a mortality rate of 0%.The cure rate was 100%in the middle-dose administration group and 33.3% in the high-dose administration group.This shows that the dosage of the monomer and the therapeutic effect were not concentrationdependent.The effect was best when the dosage is 6 mg/kg.
