The experiment made Isatis indigotica and seedings as the materials, then explored the effection of different origins、different K levels and K species on the growth、active ingredient content of Isatis indigotica and so on. We should choose a suitable K level、 K specials and develop an appropriate seed quality standards. The results of the experiment are as follows:1.The experiment measured purities,weight, water content, vigor and germination rate of isatis indigotica seedings 30 places. The results showed that:the seed purities were measured by "unarmed analysis"; the moisture was determined at 105℃ for 2 h;the vigor was determined by 0.3% TTC for 15 h;a piece of paper for seed germination,the best germination temperature was 20℃, germination time for counting was 10 d.Measured the purity,moisture content,weight, viability and germination of 30 seedings from different regions germination rate.Analysed through K cluster. the seeds were separated into three levels, the first level:germination rate≥84%, the moisture content≤ 7.63%, weight per 1000 seeds≥10.29 g, purities≥84.84%, vitality≥88.53%; second level: germination rate≥51%, the moisture content≤8.01%, weight per 1000 seeds≥7.0634 g, purities≥71.34%, vitality≥66.00%; tertiary germination rate≥15%, the moisture content≤8.82%, weight per 1000 seeds≥5.9786 g, purities≥47.38%, vitality≥26.22%.2. The experiment set 6 different levels of potassium (potassium dosage was K0= 0 g·kg-1, K1= 1 g·kg-1, K2= 2 g·kg-1, K3= 4 g·kg-1, K4= 9 g·kg-1, K5=13g·kg-1), and measured antioxidant enzyme activity,photosynthetic characteristics, active ingredients and mineral nutrients of isatis indigotica,the results showed:as the K level increasing,CAT activity increased, POD activity decreased,chlorophyll content and photosynthetic capacity improved,compared with others, K4 treatment worked best.While the K level increased,the epigoitrin and indirubin content increased before and then reduced. Phosphorus content reached highest under K4 treatment; K5 treatment is good for potassium content and Na content in the underground.Measured the growth, active ingredients(Indigo,Indirubin and epigoitrin) content and the mineral element content under three K specials.The results showed:the root length bigger and Mg、Ca content of aboveground higher under KCl and KNO3 treatment; KNO3, KCl and K2SO4 can improve the Ca content in the root and inhibit the Mg content; KNO3 improve Fe, Al and Na content underground of isatis indigotica, K2SO4 was in contrast;K inhibited the accumulation of Fe and Al, but improved the Na content,the potency was KNO3<KCl<K2SO4.3. Drought stress affect the growth、physiology and biochemistry of the isatis indigotica.Water stress decreased chlorophyll content and the photosynthesis,at the same time,the active ingredient indigo, indirubin and epigoitrin decreased. K fertilizer significantly alleviated the effection of drought stress.The biomass accumulation, SOD, CAT,POD activities, soluble sugar and proline content were all increased, the photosynthetic pigmentsthe increased before and then decreased. Drought and K fertilization were both good for the accumulation of indigo, indirubin and epigoitrin.4. When isatis indigotica was under NaCl stress, biomass reduced but root-shoot ratio increased, while the SOD, POD, CAT activities,soluble protein, proline and active ingredients(indigo,Indirubin and epigoitrin) content were all significantly increased.But,chlorophyll,carotenoid content and Pn were significantly reduced. K fertilization can promote the accumulation of biomass and the activities of autioxidant enzyme(SOD, POD, CAT); photosynthetic capacity, soluble protein and soluble sugar content improved under the K treatment,K treatment also can promote the accumulation of active ingredients(indigo,Indirubin and epigoitrin),but were bad for the accumulation of proline and total free amino acid.
