Yunnan and Beijing Isatis indigotica were used as research materials to study the vernalization characteristics and cold resistance of Isatis indigotica in order to provide scientific basis for the high-quality Isatis indigotica varieties selection. And genetic diversity and the law of effective composition accumulation were studied by using Chinese-cabbage-leaf, cabbage-leaf, mustard-leaf and tetraploid Isatis indigotica to provide scientific basis for the production and quality control of Isatis indigotica varieties. The main results were summed as follows:1. The6leaf stage of seedlings of Yunnan and Beijing Isatis indigotica were vernalized at4℃for25days, and the metabolism indicators of carbon and nitrogen were assayed. The results showed that the soluble sugar content, reducing sugar content and soluble protein content increased after vernalization and rose significantly near vernalization finishment. However, starch and total nitrogen concentration decreased significantly and reached the lowest after vernalization finishment. It is found by the analysis that high C/N value can promote the low temperature vernalization of Isatis indigotica.2. The experiment studied the effects of chilling stress on Autioxidant enzyme activities, MDA contents and soluble sugar contents by using Yunnan and Beijing Isatis indigotica seedlings for materials. The results showed that the activities of SOD, POD and CAT in two varieties were enhanced with the temperature decreased, and the increase amplitude of POD was highest, CAT took the second place. MDA content and soluble sugar content both increased with the temperature decreased. The increase amplitude of Yunnan Isatis indigotica in MDA content was higher than the increase amplitude of Beijing Isatis indigotica. The increase amplitude of Beijing Isatis indigotica in soluble sugar content was higher than the increase amplitude of Yunnan Isatis indigotica.3. It was discovered that there was significant differences among four different varieties in leaf and petiole cyanine glycosides color of Isatis indigotica intensity by researching the morphologicalmarker, pollen electron microscope scanning and molecular marker of Chinese-cabbage-leaf, cabbage-leaf, mustard-leaf and tetraploid Isatis indigotica. We find that the pollen grain of existence and surface ornamentation is a common characteristic of different varieties of Isatis indigotica by observing the pollen size, shape and surface ornamentation, etc. The form of pollen size and mesh, reflects the different varieties of differences can be a reference for germplasm diversity of Isatis indigotica. The analysis found that in terms of identifying of Isatis indigotica ITS2sequences length was shorter, easier to amplification, primers better generality and the appraisal results are consistent based on studying four different varieties of Isatis indigotica ITS2fragment.4. The contents of indigo and indirubin in Leaf and leaf in different parts of Chinese-cabbage-leaf, cabbage-leaf, mustard-leaf and tetraploid Isatis indigotica were determined by HPLC. The results showed that there were significant differences among four varieties in contents of indigo and indirubin. The indigo content in mustard-leaf Isatis indigotica was highest and the indirubin content in cabbage-leaf Isatis indigotica was highest.5. The contents of epigoitrin in tap root and lateral root of the root of Chinese-cabbage-leaf, cabbage-leaf, mustard-leaf and tetraploid Isatis indigotica were determined and epigoitrin contents in xyiem and phloem were compared. The results showed that the difference in epigoitrin contents between tap root and lateral root was significant. Contents of epigoitrin of four vareities in tap root were higher than that in lateral root.The difference in epigoitrin contents between xyiem and phloem was significant. Epigoitrin contents of four varities in phloem were higher than that in xyiem.
