Edible fungus was able to form fruiting bodies or sclerotic organization of large edible meat (or glial) the general term of the higher fungi. Because of delicious and rich-nutritional, edible fungus was more and more accepted. Through the study on the plate culture of mycelium of Agaricus bisporus by adding lead and selenium respectively, this paper studied on lead and selenium enrichment in response to growth of mycelium of A. bisporus. By A. bisporus cultivation experiment adding different concentrations of selenium, this paper studied the changes of nutrient content, physiological and biochemical properties and antioxidant enzyme system, as well as selenium form of the distribution in fruiting bodies of A. bisporus for effect and mechanism of selenium on the growth of A. bisporus and the application of selenium in the edible fungus products.The main results of research were as follows:1. This article first studied on the growth and accumulation response of Se on A. bisporus. As the result, in the range of tested concentration, When Se was 20 mg/kg and 40 mg/kg, accumulation of Se could be 62.7 mg/L and 131.4 mg/L; when Se was 80 mg/kg and 160 mg/kg, Se significantly inhibited the growth of mycelium, growth inhibition rate reached respectively 14.4% and 63.9%, accumulation of Se could be 193 mg/L and 339 mg/L.2. This paper studied the effects on physiological and biochemical by selenium-rich cultivation in fruiting bodies of A. bisporus. The results showed, in low and medium concentrations (20-40 mg/kg) of Se, the soluble protein and total sugar content increased and decreased in high concentrations (Se≥80 mg/kg), while the free amino acids showed the opposite phenomenon. With the increase of metals concentrations, the catalase (CAT) activity was decreased first and then increased while the superoxide dismutase (SOD) and malondialdehyde (MDA) content showed opposite. These result showed, low and medium concentration selenium-rich cultivation in fruiting bodies of A. bisporus could increase nutritional quality and enhance antioxidant ability while it was opposite when high concentration cultivation.3. The various forms of selenium in selenium-enriched A. bisporus fruiting bodies would be separated and determinated. Through extracting by organic solvent, organic components and inorganic components of selenium-enriched A. bisporus fruiting bodies were separated, when Se was 20 mg/kg the organic most could be 50%. Then through the crude protein and polysaccharide extraction method, Se-protein and Se-polysaccharide would be separated and determinated. In the test concentration range, Se-protein in organic could be 77-87%, Se-polysaccharide could be 5.3~45.5%. Se-protein was the main carrier of organic components.
