Pueraria lobata (Willd.) Ohwi is a genus of leguminous papilionaceae, distributed in most provinces of China, and distributed in Shanxi Province from south to north.Pueraria lobata is medicinal and edible plants, the active ingredient exist in the whole plant, but the root has the highest content of active ingredients. Its root is prescribed in "People’s Republic of China Pharmacopoeia" as medicinal material. It contains isoflavones, triterpenoids as the main active components, which can expand cardiovascular system, relieve cardiac excitability, relieve heart disorders and other effects. Its root can also be made into powder, drinks, jelly, bread, oral and so on. Its stems rich in fiber, can be made cloth and rope. Its flowers also have isoflavones, which can be used to alleviate the metabolic abnormalities caused by alcohol. Its leaves rich in chlorophyll, carotene, vitamins, minerals, flavonoids, glycosides and others, which can be used for the food coloring and fodder.Increasing people recognize for its medical and health effects, the demand of Kudzu also gradually increases in market. However, Kudzu sold on the market almost entirely from field collection. Kudzu resources diminish because of excessive excavation and changes in the ecological environment. It has been difficult to see large distribution groups. Therefore, tending wild plants and cultivation will gradually become the mainstream of medical supply of Pueraria lobata.Pueraria lobata is lianas, in the process of cultivation, it often adversely affect the vicinal crop because of disorderly growth and rapid spread of the stems. The small proportion of roots is due to the vegetative organs of stems and leaves is too much. The main parts of the useful component is present in the phloem, but the proportion of phloem in anatomy is not high. Therefore, digging wild plants resources, establishing wild tending technology, directional selecting and fostering cultivation varieties, these has become kudzu medicine production problems to be solved.(1) In this experiment, using the Pueraria lobata collected from six different places of Shanxi province as raw materials, establishing a rapid propagation method of its test-tube plantlets, then analysis and compare the growth of roots, stems and leaves of different places of the test-tube plantlets, which provide an important basis for the subsequent screening.The experiment results in following conclusions: ①using a clean and dry brush to remove surface dust,75% ethanol disinfecting for 90 s and 0.1% HgCl2 solution disinfecting for 10 min, this is the best disinfection conditions. The MS medium of 6-BA-1.0 mg·L-1, KT-0.3 mg·L-1 is suitable for primary culture, the MS medium of 6-BA-1.0 mg·L-1 is suitable for subculture, thel/2MS medium of I-BA-1.0 mg·L-1, DSC-0.5 mg·L-1 is suitable for rooting. ② There are some differences in growing of roots, stems, leaves of different place of Pueraria lobata.(2)This study established an effective method of extraction and determination of chlorophyll of Pueraria lobata, and also compared the content of chlorophyll of wild plants and test-tube plants chlorophyll. This provides the basis for the use of Pueraria lobata leaves and selection of germplasm.The experiment results in following conclusions:the leaves’chlorophyll content of different origin wild plants or test-tube plants is different, this illustrates that different origin have different physical characteristics. However the differences trend of chlorophyll content in wild and test-tube is approximately the same. This indicates that test-tube plants retain the genetic characteristics of the original wild plants.(3) In this experiment, using RAPD technique to compare genetic relationships of different places of test-tube plantlets from the molecular level. It is revealing genetic background and relationship in resources in Pueraria lobata of Shanxi. It also provided a basis for mining, conservation, development and utilization of Pueraria lobata germplasm resources in molecular biology.The experiment results in following conclusions:six different origin Pueraria lobata can be divided into three categories. The Pueraria lobata from Hou-gou-kou, Ping-lu and Ling-chuan belong to Group Ⅰ. The Pueraria lobata from Xiao-bei-shan belong to Group Ⅱ.The Pueraria lobata from Yan-hu and Ling-qiu belong to Group Ⅲ.As so far, it not been seen the reports about rapid propagation, determination of chlorophyll content and molecular marker analysis of Pueraria lobata of Shanxi yet.
