Pueraria lobata is the underground root of Pueraria lobata,a perennial vine,rich in starch and isoflavones of Pueraria lobata.It is the first medicinal and food homologous plant approved by the Ministry of Health of China and has great medicinal and edible value.For a long time,the varieties planted in Pueraria lobata production area are relatively single.Long-term continuous cropping and asexual reproduction have seriously degraded the original fine germplasm,caused frequent diseases,reduced yield and quality.These problems can be effectively solved by plant tissue culture,purification,rejuvenation and virus-free technology,but the low survival rate of plantlets restricts the promotion and application of tissue culture virus-free seedlings.In this study,Guige No.1,a new variety of Pueraria lobata,was treated with different sucrose concentration,shading,seedling-raising substrates and water-retaining agents.The effects of different environmental conditions on the survival rate,growth characteristics,yield and quality of Pueraria lobata plantlets were evaluated comprehensively.The key factors restricting the survival rate of Pueraria lobata plantlets and high-yield cultivation techniques were discussed,which could provide reference for the establishment of high-efficiency cultivation techniques system regulations and the promotion of improved varieties of Pueraria lobata plantlets.The results are as follows:1.The effect of sucrose concentration on the growth of plantlet and transplanted seedlings in shed ranged from big to small:4%>6%>5%>3%.The survival rate of plantlets in shed and field ranged from big to small:4%>3%>5%>6%.The highest survival rate was 4%sucrose,86.7%and the lowest was 6%sucrose,68%.The difference was significant.2.The survival rate of transplanting under shading condition was 84%,and the survival rate of non shading control was 53%.Under shading condition,the survival rate of root transplant seedlings of Pueraria lobata increased by 31%compared with non shading control.The difference is significant.3.The order of survival rate of plantlet in greenhouse and field transplantation from large to small was:matrix Ⅰ>matrix Ⅴ>matrix Ⅱ>matrixⅣ>CK>matrix Ⅲ,and the highest survival rate of matrix Ⅰ was 93.2%and that of control was 65.13%.After transplantation,the promotion of plantlet growth in shed and field was significant.The order of effects ranged from large to small:Matrix Ⅰ>matrix Ⅳ>matrix Ⅱ>matrix Ⅴ>matrix Ⅲ>CK.The order of effect on average plant weight and yield of plantlets was matrix Ⅰ>matrix Ⅱ>matrix Ⅴ>matrix Ⅳ>CK>matrix Ⅲ.The order of comprehensive evaluation of quality was matrix Ⅰ>matrix Ⅳ>matrix Ⅱ>matrix Ⅴ>matrixⅢ>CK.4.The effects of water-retaining agent treatment on the survival rate of shed seedlings and field transplantation,the growth promoting effect of plantlet sand the quality of tuber roots were as follows:dipping roots and irrigating roots>irrigating roots>dipping roots>CK.
