Analytical instruments as high-performance liquid chromatography(HPLC) were adopted to detect the content of dihydromyricetin and related flavonols in Ampelopsis grossedentata(Hand.-Mazz.) W.T.Wang, and the SPSS22.0 software was used to analyze the correlation and illuminate the key precursors and competitive substrates involved in the biosynthetic pathway of dihydromyricetin.1) The method to detect the content of dihydromyricetin in A. grossedentata was established, and the linear regression equation could be stated as follows: Y=28327X-101.2(R2=0.996), with a limit of quantity(LOQ) at 35.74 ng, which showed a good linear relationship in the range of 0.5μg~10.0μg.2) A way on simultaneous analysis of dihydromyricetin and myricetin by the HPLC-UV method has been established, which has good stability, reproducibility and precision.3) To analyze the eleven flavonols including dihydromyricetin, myricetin and quercetin simultaneously, PTLC-HPLC method has been established.4) The application of simple UV-Vis spectrophotometry technique was for the detection of the total flavonoids in A. grossedentata, which could provide an effective technical reference for the appraisal of A. grossedentata.The research will provide reference for further study on the molecular mechanism of the metabolic accumulation of dihydromyricetin in A. grossedentata.
