Paeonia [Paeonia rockii(SG Haw et LA Lauener)] belongs to Paeoniaceae Paeonia Moutan, is native to China’s wild peony and important medicinal plants, which has a long history of cultivation that is widely used in garden greening and landscaping with high ornamental value. In order to chose the best explant and medium, establish a reliable and efficient system of callus induction. In this paper, a study of the effect on callus culture by using different culture medium and growth regulators with different concentrations was proposed, which used vegetative organs and reproductive organs of Paeonia rockii as materials. The main results showed that:1. The best sterilization time respectively for leaves, petioles were 6 min, 8 min.2. The best medium for embryo germination was B5+6-BA2.5mg/L+CH0.5g/L+Sugar20g/L+ AGAR5g/L.3. The ability to callus induction of Vegetative organs from high to low was: sterile petioles> sterile leaf>field petiole>field leaf. The best explant was sterile petiole. The best medium was MS+2.0 mg/L6-BA+0.4mg/L NAA+LH0.5g/L+PVP0.1g/L +Sugar20g/L+AGAR5g/L.4. The ability to callus induction of Reproductive organs from high to low was hypocotyl> cotyledon>petal>stamen>carpel>stigma>filament>anther. The best explant was hypocotyl. The best medium was MS+2.0mg/L6-BA+1.0mg/LNAA+2.0mg/L2,4-D+LH0.5g/L+PVP0.1g/L+Sugar20g/L +AGAR5g/L.5. The callus induction rate from outer to inner of the morphological structure of Paeonia rockii organs was gradually reduced(petal>stamen>carpel>stigma) and browning rate was increasing(petal< anther<stigma<carpel).6. The best proliferation medium for sterile petiole was MS+2.0mg/L2,4-D+1.5mg/LNAA+CH 0.5g/L+Sugar20g/L+AGAR5g/L. The best proliferation medium for hypocotyl wass B5+0.6mg/L6-BA+LH0.5g/L+PVP0.4g/L+Sugar20g/L+AGAR5g/L.
