Superovulation And Collect Oocytes By Surgery In Dogs

The main content of this experiment was divided into three parts, the first part was the superovulation in dogs, the second part was to obtain mature oocytes by surgery, the third part was to observed oocytes by microscopy in the lab.The superovulation of dog was performed under the condition of injection PMSG and hCG, chose 5 healthy and diestrous bitches(weight:about 5 kg), the first day of the experiment was to inject the PMSG to the bitches by continuous injection for 8 days at a dosage of 200 IU/d. the 11th day injected the hCG at a dosage of 2000 IU, the 21th day to collect the oocytes by the surgery.Surgery of collectting oocytes was based on the superovulation of bitches, the bitches were anesthetized with the zoletil 50, along midline of abdomen behind the belly,made a skin incision, cut the skin and subcutaneous tissue, abdomen was opened with median incision, find and removed the uterus, ovary and oviduct, then brought them to the lab with vacuum flask.Before of observing oocytes by microscopy in the lab, washed the uterus, ovary and oviduct, cut the redundant tissue, flushed(10ml liquid) with the needle of 1 ml from the junction of the uterus and oviduct, on the other end use the plate received the flow of the liquid. Use hyaluronidase mechanically strip of cumulus cells and obtain oocytes, and regarded the oocyte with pb I as the mature oocyte. Used the cell dyeing method we can see the pb I clearly. Results: We obtained the mature oocytes successfully.This experiment was aimed to obtain mature oocytes through the method of dogs’ superovulation which was injected PMSG and HCG during non-breeding season (on January).
