Vitis davidii Foex distributed widely with multi-resistance and good agronomic traits is a kind of wild grapes native to China. Based on the analysis of morphological, palynology and ISSR markers of25germplasm resourses of V. davidii Foex in Hunan, Jiangxi and Fujian, we aimed to reveal the morphological, palynology and genetic diversity, and provide theoretical for the protection and research on wild V. davidii Foex. This study aslo analysed the morphological, palynology and ISSR markers mathed. The main results as follows:(1)The variation coefficients of the16morphological characters from4.98%to28.57%, the principal component cumulative contribution rate of83.168%, the result of morphological analysis showed that there were high phenotypic diversities within the populations. The populations can be divided into three groups by using UPGMA method.(2)Based on the evolution of the pollen grains from big to small order, the populations can be divided into three groups. Based on the evolution trend of the pollen, the polar surface of the pollen evolve from cavernous to mesh to verrucous.(3)Efficient method of total DNA extraction from grape was developed. The ISSR-PCR reaction system was optimized. The ISSR-PCR amplified reaction was carried out13primers from100arbitrary primers. The result was:139polymorphism bands among116amplified bands. The populations can be divided into four groups.(4)Phenotypic traits largely consistent with the results of ISSR molecular markers. Morphological analysis can be intuitive judgment difference, but the need to quantify the value of the statistics and calculation are different. Palynology can reveal the evolution of the populations and can distinguish between morphological differences grape species. ISSR molecular markers from the perspective of genetic research related, more accurate than the formers, but it can’t represent. Morphology, palynology and ISSR marker can also show that there were high phenotypic diversities within the populations.
