| Firstly we analyzed the KISS-1gene bioinformatics in this study, made a prediction about physical and chemical properties of the gene encoding the product, the structure and the function, built phylogenetic trees, then four swine breeds including two exotic breeds of Landrace, Large White and two Chinese local breeds of Shaziling, Daweizi pigs, totally353pigs were used as experimental materials to study the KISS-1gene single nucleotide polymorphisms. We used PCR-RFLP for the genetic polymorphisms analysis, and associated three variable sites and3porcine reproductive traits. The main results are as following:Bioinformatics analysis shows that KISS-1gene coding product is a hydrophobic protein, not a trans-membrane protein. Further analysis shows that the protein have7phosphorylation sites and9glycosylation sites and disulfide bond between position53-71.Predicted signal peptide cleavage site is between amino acids17-18.The study on the gene structure discover that there is a low complexity sequence between31-42aminoacid.The encoded product plays a major role in the biological nucleus with signal transduction, hormones, transcription, transcriptional regulation function.Secondary structure of KISS-1gene is mainly encoded by a-helix and random coil. The analysis of phylogenetic tree shows that pigs have the highest homology with cats and ferrets, have the second highest homology with cattles, sheeps and humans.have the lowest homology with mus musculus.The sequenced and compared PCR products are used directly. which are combined with the PCR-RFLP method.detect that KISS-1gene Exonl-149locus C→A mutation, AA changed from V to F, and Exon2-198points G→C mutations. AA changed from E to Q,and Exon2-294locus A→G mutation, AA changed from A to G. The locus of three sites which are gene Exonl-149C→A, Exon2-198G→C. Exon2-294A→G. in the distribution of four different breeds is detected, we found that local breeds homozygosity are generally lower than foreign breeds, but polymorphic information content is richer than foreign breeds.Through association analysis of KISS-1gene polymorphisms and some reproductive traits.we found that Yorkshire Exon2-294A→G mutation affect TNB and NBA.The dominant genotype is AA, compared to GG and AG, there is a significant difference.The TNB is1.77and2.02higher respectively (P<0.05), and the NBA is1.83and1.22higher respectively (P<0.05). What need further validation is whether this will improve the fertility as genetic markers.The result of this study shows that the The KISS-1gene may be used as a molecular genetic markers of pig reproductive traits.Through further bioinformatics exploration, it will have a very important significance on the study of KISS-1gene and its encoded protein function. |
