As the quantitative traits and economic traits, Porcine reproductive traits’ heritability especially is low. For Chinese local breed swine it is better than others.The combination of traditional hybridization and Marker-Assisted selection become a new way of breeding and its protection. In this paper, Jiaxing black porcine was selected as research sbujects,we select related major genes(estrogen receptor, follicle- stimulating hormone β receptor, prolactin receptor) and we choose 513 ear tissues of Jiaxing black pigs as experiment subject and isolate genomic DNA from ear tissues. By the candidated genes’ exon region we design primers. By PCR technology we obtain the purpose sequence. Detected SNPs with PCR-SSCP method and directly Sequencing. We statistically analyze the gene allele frequency and genotype frequency, Hardy Weinberg equilibrium condition and correlation analysis results with porcine reproductive traits, which have done contribution for established molecular breeding pattern of MAS of Jiaxing black porcine.The ESR gene in the first exon area amplified purpose fragments.Sequencing analysis find four SNPs: T6901 C, G6759 A, T6927 G, A7188 G. The peak figure of fragment each SNP appeared three genotypes, respectively named AA, AB, and BB, NN, NM, MM, EE, EF, FF, JJ, HJ and HH. We statistically analyze the gene allele frequency and genotype frequency.χ2 test showed that T6901 C, A7188 G are in Hardy Weinberg equilibrium, G6759 A, T6927 G significantly deviate from the Hardy Weinberg equilibrium state. In this study we Select 5 reproductive traits of jiaxing black pig, consisting of Total number of piglet born, number of piglet born alive, number of stillborn pigs, farrawing interal, litter weight at birth. T6901 C and A7188 G with six genotypes correlation analysis results show that Jiaxing black pig association analysis found that it is no significant difference with T1069 C site. While it is in A7188 G site HH genotype for litter size was significant higher than JJ genotype(P<0.05). For the number of stillbirth traits HH genotype was significantly higher than JJ genotype(P<0.05). For litter weight at birth it is significantly higher that HH character traits is than JJ genotype(P<0.05).For other traits it is no significant difference by correlation analysis.FSH beta gene in the exon 2 by sequencing received three SNPs: T1035 C, A922 C, A925 G. every site has three genotypes, respectively named KK, KL, LL, QQ, QR, RR, UU, UV, VV. We statistically analyze the gene allele frequency and genotype frequency.χ2 test show that A922 C site significantly deviate from the Hardy Weinberg equilibrium, T1035 C, A925 G site comply with Hardy Weinberg equilibrium state. The genotypes of T1035 C site and A925 G site with 5 reproductive traits of jiaxing black pig continue correlation analysis, the results show that T1035 C site was not found significant difference with five reproductive traits. G925 A site found by correlation analysis UU genotype and Vvgenotype in LWB was significantly higher than UVgenotype(P < 0.05).PRLR gene in exon 5 area found A SNPs: A13902 T, By PCR-SSCP it appears three genotypes: XX, XY, and YY. We statistically analyze the gene allele frequency and genotype frequency.χ2 test showed that the site in accordance with Hardy- Weinberg equilibrium; After association analysis with 5 reproductive traits of jiaxing black pig, results show litter weight at birth, the number of stillbirth traits and litter size show that XX, XY genotype, YY genotype was significant difference(P < 0.01). the other traits were not found.A7188G site and A13902 T site have combined genotype effect about five reproductive traits’ association analysis, which show that for litter size HHXX genotype is higer than JJXX genotype(2.03 piglets born per litter,P<0.05), HJXX genotype is higher than JJXX genotype(2.12 piglets born per litter,P<0.05),HJXX genotype with TNB is more1.95 piglets born per litter than JJYY(P<0.05). For the NBA trait, HHXX genotype is higher than JJXX genotype(1.95 piglets born per litter,P<0.05),it is higher than JJYY genotype(3.53 piglets born per litter,P<0.03). HHXX genotype is more 4.37 kg per litter than JJYY genotype about LWB, it is higher than JJXY genotype(3.83 kg per litter,P<0.05). There is no significant difference about Number of stillborn pigs and Farrawing interal.
