In this paper, the ELISA and real-time PCR were applied to investigate and analyze porcinepseudorabies in large scale pig farm in Tianjin pig industry from2011to2013.Sera were collected from29large scale pig farms in8counties of Tianjin. In ELISA test, serum gBantibody positive rate was84.94%. It rises gradually from78.56%,86.06%to90.20%in the year of2011,2012and2013respectively. Serum gE antibody positive rate was26.41%. It declined year byyear from34.77%,28.86%to15.61%. The results showed that the PR virus infection reduced graduallyin the field. The antibody positive rate of gB and gE were different in different regions. The boars, sows,piglets and fattening pigs were analyzed for detection of gB antibody and gE antibody in3years. ThegB antibody level in piglets maintained the highest (except2013sows). The gE antibody positive ratewas the highest in boars, that should be strengthened the detection and eradication. On the detection ofserum results according to the seasonal analysis shows, the second season was the peak of porcinepseudorabies occurrence.By using the method of gE-ELISA, the wild virus infection status was detected in pigs inoculatedwith domestic and import vaccines. The wild virus infection rate were31.60%,20.68%and13.29%from2011to2013in the domestic vaccine herd, and that were22.23%,14.40%and8.31%in theimport vaccine herd. The positive rate was decreased gradually with both vaccines. However, virusinfection rate in domestic vaccine herd was higher than that in import vaccine herd. Immune efficacy ofimport vaccine was obviously better than domestic vaccine.Clinical samples from18pig farms of TianJin from2011to2013were tested by real time PCR.The positive rate of PRV was12%. The positive samples were detected with gE real time PCR for wildvirus infection. The gE positive was13.91%of the number of PRV positive samples and1.64%of totaltested samples. gE positive rate decreased annually from19.64%in2011to14.12%in2012, and to7.62%in2013. Those results were consistent with that of ELISA. The results were analyzed accordingto region; it was coinciding with ELISA due to small number of samples. The real time PCR resultsshowed, as in ELISA, that the second quarter is the main epidemic season and wild virus infection ratewere higher in boar and piglet. The results were consistent with that of ELISA.
