| In this study, the epidemic survey of foot-and-mouth disease had been investigated by methods of Multi-RT-PCR and 3ABC-I-ELISA. 104 Samples of catlle and 386 Samples of pig collected from 4 areas (QingYang DingXi QingShui JingTai) of gansu province were detected in the light of Multi-RT-PCR kit and 3ABC-I-ELISA kit in etiology and serology. Non-Structure protein of animals antibody were detected through NS-3ABC protein antigen to identify if the local animals had ever been infected by FMDV, which can make a reasonable inference for the foot-and-mouth disease epidemiology of local animals. Through serology test. we found that one of 104 cattles, serums NS-3ABC was positive and two (QingShui)were suspicious. It,s positive rate was 2.4%. And we found that two pigs, (DingXi) serums NS- 3ABC were positive and it,s positive rate was 2.1%. At the same time, 6 cattle,s O/P were positive through Multi-RT-PCR. The main immunogen coding gene VP1 of FMDV was amplified and sequenced to draw the phylogenetic tree. Some refencence strains used here in order to compare with our target sequences. Thus it appeared that the partial VP1 gene sequence of 6 FMDV strains from cattles, O/P fluid were type O. And the evolution branch were near to O/JS/08-1. Although most of animals were nagetive,a few of positive ones should be noticed. |
