With the extensive application of antimicrobial agents in animal husbandry, not onlydrug-resistant strains are becoming more and drug-resistant spectrum becomes morewidely, but also brought serious challenges to the animal food safety. So the research anddevelopment of green, no residue of antibacterial, antiviral herbs will have broad prospectof application in the veterinary drug production. The purpose of this research is to study onthe best water extracting procedure of Jiegeng tang (Fago40g, Houttuynia25g, Platycodon8g, Licorice8g) for future industrial production, and evaluation of preventive effect onpulmonary edema in mice.The content of this research includes several aspects:1. Identifying the Chinese herbal medicine of Platycodon, licorice, Fago and Houttuynia.The method used are physical and chemical identification, microstructure identificationand characters identification, the results are in line with "Chinese VeterinaryPharmacopoeia" standards.2. Implement in vitro antibacterial test with the water extract of the four kinds of singleherb and the self prescription. Methods: chose E.coli, Pseudomonas aeruginosa,Staphylococcus aureus and Haemophilus paragallinarum these common bacteria, methodof Oxford cup was used to test the bacteriostatic ring and solid medium serial dilution wasused to test the minimum inhibitory concentration. Results: the solid medium serialdilution test showed that, all the extracts proved have antibacterial activity to all pathogens,and the minimal inhibitory concentration of the self prescription was lower than others.The Oxford cup method showed that, the sensitivity of Fagopyrum dibotrys(D.Don)Haraand Houttuynia cordata to E.coli and Pseudomonas aeruginosa were low, but the sensitivityof the self prescription to those bacteria was relatively high.3. Considering the soaking time, solid-liquid ratio, extraction time and extraction timeswill influence the water extraction process, so using the L9(34) orthogonal test tablemethod, comprehensive analysis the yield of extracts, licorice acid and rutin content tochoose the best process. Results: the best process was12times of water, immerse2hours,decoction for3times,2hour for the first time. The second10times of water, decoction1.5hours, the third time8times of water, decoction1hours.4. The mice in test were divided into control group, pathologic model group andpreventive group, preventive group was lavaged flavored Jiegeng Tang in the morning andat night for seven consecutive days, but the remaining two groups were lavaged saline, the dose was1mL/100g. After7days, the mice of pathological model group and theprevention group induced pulmonary edema by tail intravenous injection oleic acid.2dayslater, all the mice were sacrificed, observed pathological changes of lung, comparisonbetween the three groups of lung coefficient, lung water content and lung wet weight andlung dry weight ratio, in addition, made pathological sections of lungs. Results:flavored Jiegeng Tang pulmonary edema in mice had a good preventive effect, significantdifferences between the groups.Therefore, the flavored Jiegeng Tang has a good antibacterial effect, the optimum waterextraction process is determined, and is able to play a preventive effect to pulmonaryedema, may provide some basis for clinical application.
