Abstract/Summary:
Dendrobium(Dendrobium candidum Wall ex Lindl.) is a orchid perennial herb with unique medicinal value. Under natural conditions germination rate and survival rate of dendrobium is extremely low for its living conditions is diffucult to reach; furthermore, artificial mining causes a shortage of natural resources. It is also very difficult to cultivate large-scaled even under aseptic conditions, so its market supply is serious inadequacy. At present, the artificial seed of Dendrobium has been studied a lot, but artificial seeds used for natural cultivation may meet a lot of problems, such as storage, inconvenient transportation, which have affected its mass production. In this experiment, its germination rate and survival rate are improved by coating its artificial seeds, which make it more antibacterial, water retention under natural conditions.Artificial seeds of Dendrobium are made as propagation body by sprouting protocorm or not, discussing the influence of36kinds of endosperm formula which are designed by sucrose, starch, ABA, NAA, activated carbon, carbendazim, alginate on Dendrobium synthetic seed germination, and studying the common law of artificial seed production and germination process, and its storage and preservation,to screen of suitable breeding and endosperm formula. The results show that the germination of artificial seeds is influenced not only by the seed coat composition of CaCl2and the reaction time, but also the location where the embedding pill is and status of reproductive body itself. In this case, the germination rate and survival rate of artificial seed are higher when germination protocorm is used as the propagation. The main factor to influence the germination of Dendrobium synthetic seed is sucrose and activated carbon. Appropriate treatment combination is like this: sucrose3%, ABA lmg/L, activated carbon0.5%, MBC3%, and concentration of sodium alginate3%. The longer the storage time is, the lower the germination rate is. The germination rate and survival rate of artificial seeds can be improved when antibacterial agent is added to the seed coat composition.In this experiment, on the basis of selecting the appropriate propagule and endosperm formula, an anti-bacterial membrane was designed as the outer coating of the artificial seed of Dendrobium. It is showed the germination rate and survival rate are higher than that without coating in germination and storage experiment under natural conditions.
