Horse industry is an ancient industry. China is a traditional horse country, and has a long history of horse raising. Mongolian horse has the largest number and the most widely distribution in our country, it has characteristics of amazing stamina, drought, cold, disease resistance.Mongolian horse is an extremely valuable gene pool on the horse genetic resources. Therefore, China carried out research on genes related to excellent local varieties of Mongolian horse blood glucose. It not only can further understand the causes of the strong endurance of Mongolian horse, provide a theoretical basis for revealing the durable genetic characteristics of Mongolian horse, but it also have an important significance to improvement and cultivation of local breed horse. In this study, Mongolian horse had been the research object, the physiological and biochemical index of blood of Mongolian horse were determined and analyzed which the physiological and biochemical index of blood of Mongolian horse existence differences before and after training.The results show that, after training, all the glucose, creatine kinase, urea, hemoglobin, red blood cells, white blood cells, lymphocytes and platelet count showed significant changes. Before training, the horse for blood glucose concentration is (4.77±0.42) mmol/L, after training the horse for blood glucose concentration is (5.83 ± 0.31) mmol/L, higher than before training, before and after appear significant difference (P<0.05). Secondly, we conducted transcriptome analysis of high-throughput sequencing on the muscle samples of Mongolia horse before and after training, and carried out GO Functional Enrichment to Differential Gene which were screened. Screening of LPHN3, LGALS3, IFIH1 gene is the target gene of the study in the pathway related to blood glucose. Once more, using the Quantitative Real-time PCR method tested the Mongolia horse blood and changes of LPHN3, LGALS3 in Skeletal muscle and expression of the mRNA of the IFIH1 gene before and after training. The results show that, the genes of LPHN3, LGALS3, IFIH1 in Mongolia horse are expressed in the blood and skeletal muscle before and after training. After the training, LPHN3 gene expression in the Mongolian horse skeletal muscle volume is higher than before, express the extremely significant difference (P<0.01); the expression of LPHN3 gene has no remarkable difference (P≥0.05).After training, LGALS3 amount of gene expression in the Mongolian horse blood and skeletal muscle were higher than before, the amount of expression in skeletal muscle has significant difference (P<0.05), the expression in the blood has extremely significant difference (P<0.01). After the training, IFIH1 gene expression in the Mongolian horse blood volume is higher than before the training, express the extremely significant difference (P<0.01); Before and after training, the expression in skeletal muscle has no remarkable difference(P>0.05).
