The Mongolian horse were very common in China, but they mainly produced in the Inner Mongolian and outer Mongolian Because of the special artificial breeding methods, created the Mongolian horse’s strong endurance ability. They can run for more than 100km at a top speed. This super endurance trait is determined by genetic and environments. Therefore, in order to reveal the genetic mechanism of the formation of endurance, this research choose 6 Mongolia horses which had similar genetic background, had been trained for 4 months. Then, collected muscle samples before and after the training respectively, the transcriptome analyses were carried out on skeletal muscle of Mongolian horse.Through above steps we found there is a direct relationship between MyoG gene and muscle development. Then, the expression level analysis were carried out. Whereafter, we did muscle cells culture, the original generation of culture and subculture were followed,the total RNA was extracted at the most vigorous growth period. We utilizing the RT-PCR technology testing the expression level of MyoG in Mongolian horse normal muscle and cell culturing. It was found that the expression of MyoG was significantly increased in the muscle tissue after a endurance training. However, there was no significant change between cultured muscle cell and normal muscle tissue.
