Nerium indicum Mill subordinates to the genus of Gentianales,Apocynaceae,Nerium,andit is a sort of evergreen undershrub. Nerium indicum Mill is an important species of trees forhedge with high application value. The thesis studies on the tissue culture of Nerium indicumMill, in two ways that axillary buds induction and callus differentiation to get explants, then topropagate rapidly. After that we can not only obtain a large number of in vitro non-toxic plantlets,but also provide reference technique for the research on the tissue culture of other plants inApocynaceae. Adopting Uniform Design for the experiment, it will reduce the time of selectingformulas, moreover reduce work load for us. The conclusion was made as follows:1. The stems、stem tips and leaves were used as explants, MS+6-BA1.5mg/L+NAA0.05mg/L was used as callus induction medium, and MS+6-BA1.5mg/L+NAA0.05mg/L was used ascallus differentiation medium, to choose the best explants. The results showed that the rate ofleaves callus induction was94.64%, and the rate of callus differentiation was88.89%, they wereall higher than the rate of stem tips and stems. So the leaves were the best explants.2. Adopting U10(103) Uniform Design, the result of callus induction showed that6-BA、NAAand2,4-D were the greatest effects, and the interaction between NAA and2,4-D was more thanthat between6-BA and them. Therefore the best medium for callus induction wasMS+6-BA2.00mg/L+2,4-D2.00mg/L+NAA1.40mg/L.3. Adopting U10(104) Uniform Design, the result of callus differentiation showed that6-BA、KT and the interaction between them were the greatest effects. Therefore the best medium forcallus differentiation was MS+6-BA2.00mg/L+KT0.01mg/L.4. Adopting U10(102) Uniform Design, the result of axillary buds induction showed that6-BAand NAA were the greatest effects, they were more than their interaction effect. Therefore thebest medium for axillary buds induction was MS+6-BA1.40mg/L+NAA0.10mg/L。5. Adopting U10(103) Uniform Design, the result of multiplication showed that the interactionof6-BA and GA3was the greatest effects for the index of multiplication. Therefore the bestmedium for multiplication was MS+6-BA1.12mg/L+GA31.55mg/L, and the index was about4.5.6. Adopting U10(102) Uniform Design, the result of rooting induction showed that IBA andNAA were the greatest effects, and the significance was greater than the interaction of them.Therefore the best medium for rooting induction was1/2MS(macroelement)+1/4MS(ferric salt)+IBA1.00mg/L+NAA0.02mg/L, the rate of rooting was about95%.7. The culture substrate for hardening-seedling was peat soil: fine river sand: decomposingpine=1:1:2. The survival rate was about95%on the condition of choosing strong and healthyseedlings.
