Nerium indicum Mill is an evergreen shrub in the dogbane family Apocynaceae and already has a long cultivation history in our country. Because of its strong resistance to growth conditions, antitoxic and dust-remove ability, and the long flowering period, Nerium indicum Mill is a good species for landscape engineering. Nowadays, people usually use cutting propagation. However, various planting-patterns resulted in the uneven qualities in different areas. In order to ensure a common standard of cutting propagation, we studied the influence factors, carried out a series of screening tests to identify the main factors and the optimal experiment combinations. The study provided an standardization and guidance on Nerium indicum Mill’s cultivation.Firstly, in order to sift suitable materials for rooting, we selected five kinds of matrix, i.e. two composite matrix (M2and M3), two sole matrix (M4and M5) and garden soil (M1; CK), Cutted the branches into these matrix respectively, and then we inspected the physiological and biochemical indixes of the materials. Secondly, Orthogonal Experiment L9(34) was utilized to filter out the main factors which affect the Nerium indicum Mill cutting’s propagation and the optimal experiment combination. The results were shown as following:(1) According to previous research, the Nerium indicum Mill is easy to root, and the regeneration type was the combination the formations of phloem parenchyma and callus. It’s feasible to accomplish the large-scale asexual reproduction using Nerium indicum Mill’s branches.(2) Rooting and growth in different matrix are different, the experiment turned out that the complex is better. Compared with the Control (Mi) and sole matrix (M4and M5),The two mixture, M3, and M2, are suitable for plant growth, especially, the mixture of M3.(3) In adopting multifactorial experiment, we got main factors though investigating the root’s physiological biochemical character. The influences are the type of cutting, the kind of auxin and the concentration, successively; and the optimum combination is the treatment of A1B1C1, that is, using the upper branches which is lighter degree of lignification as cuttings, then the cuttings were treated with a concentration of200mg/L NAA solution for10min, the root’s growth tendency is the best compared with others.Considering comprehensively the various indicators, we have the main factors. i.e, the type of cuttings (A) and the kind of growth regulator (B), which influence the Nerium indicum Mill’s rootability, and the optimum combination is the treatment of A1B1C1, that is, using the upper branches which is lighter degree of lignification as cuttings, then the cuttings were treated with a concentration of200mg/L NAA solution, the rootability is best.
