Castanea henryi is an important fruit and timber tree species in south china,of which the ecological benefits and economic value is huge.At present,most Castanea henryi seedlings in China are propagated by methods such as grafting and seeding whose production cycle is long,so it can not meet the needs of breeding of castanea henryi seedlings.The tissue culture technology can be used annually for industrial production,and can provide a large number of high-quality Castanea henryi seedlings in a short time.Therefore,it is urgent to carry out research on tissue culture technology of Castanea henryi.In this study,we use embryos,stem segments and leaves of castanea henryi as explants to study the tissue culture of castanea henryi.The main research rusults were as follows:(1)Tissue Culture of Embryos.Ethanol 50 s + hydrargyri bichloridum 10 min had the best disinfection effect on embyros,the disinfection rate and browning rate were 12.10%and 23.33%;1.0 g/L citric acid had the best anti-browning effect on Castanea henryi embyros,and the rate of embyro browning was 8.80%;The seedling rate of embyros was the highest in the medium without plant growth regulator,which was 80.07%;The optimum medium for cotyledon callus induction was 6-BA 2.0 mg/L + NAA 1.5 mg/L,the induction rate was 76.27%.and the optimum medium for cotyledon node adventitious bud induction was 2,4-D 0.1 mg/L,and the induction rate was85.67%.(2)Tissue Culture of Stem Segment.Ethanol 30 s + hydrargyri bichloridum 10 min had the best disinfection effect on stem segment,the disinfection rate and browning rate were 13.43%and 33.33%;1.0 mg/L citric acid had the best anti-browning effect on Castanea henryi stem segment,and the rate of browning was 14.53%;6-BA 2.0 mg/L+IBA 0.5 mg/L was the best combination for axillary bud induction in shoot segment of castanea henryi with buds,and the rate of axillary bud induction was 66.67%;6-BA 1.0 mg/L + NAA 1.5 mg/L was the best combination for multiplication,the induction rate was 57.78%and the reproductive coefficient was 4.03;6-BA 2.0 mg/L + IBA 0.1 mg/L was the best combination for strength and elongation of shoots,with an average seedling height of 3.50 centimeter;the most suitable culture medium for rooting was WPM + IBA 3.0 mg/L + NAA 0.5 mg/L,and the rooting rate was 50.61%.(3)Tissue Culture of leaves.ethanol 30 s + hydrargyri bichloridum 10 min had the best disinfection effect on leaves,the disinfection rate and browning rate were 11.07%and 29.67%;1.0 mg/L citric acid had the best anti-browning effect on Castanea henryi leaves,and the rate of browning was 9.14%;The combination with the highest induction rate of leave calllus was6-BA 1.0 mg/L + 2,4-D 1.0 mg/L,and the callus induction rate was 62.79%;the best combination of plant growth regulators for callus multiplication of Castanea henryi leaves was 6-BA 1.0 mg/L+ 2,4-D 0.5 mg/L,and the multiplication coefficient was 5.56.
