Wheat is one of the most important crops in the world. However, due to transgeneticreceptor‘s regeneration ability, wheat is the last successful transgenic crops, it is still achallenge. Wheat has rich germplasm resources, There are many differences in them.Thereare376kinds of different wheat genotypes’ mature embryos as explants for tissue culture inour research. We can divided them into nine groups by wheat callus regenerativecapacity.Search for some wheat genotypes as suitable receptors of transgenic; compareindices of physiological and biochemical which are different genotypes of wheat callus,selected indicators which is relevant with wheat regeneration; using gene gun to tranformswitchgrass auxin transport-related PvPIN1genes into wheat, in order to obtain the newtransgenic wheat.The results are as follows:1. The average callus rate was89.40%of wheat mature embryo, the averageembryogenic callus rate was14.58%, the average browning callus rate was5.23%, theaverage callus differentiation rate is12.62%,the average rate of seedling regeneration ofcallus from mature embryos is5.68%.2. Embryonic callus rate and differentiation rate and seedling rate is a significantpositive correlation (P <0.01), the correlation coefficients were0.623and0.574.Embryoniccallus rate and browning rate were a significant negative correlation (P <0.01),correlationcoefficient is-0.253. Differentiation rate and seeding rate were significantly positivelycorrelated (P <0.01) with a correlation coefficient of0.76, the differentiation rate andbrowning rate showed a significant negative correlation (P <0.01),correlation coefficient is-0.337. Besides the callus rate and differentiation rate, seedling rate, browning rate irrelevant.3. Cluster analysis showed that all wheat genotypes can be divided into nine groups, ofwhich the first and second groups’callus have the best regenerative capacity;3rd,4th groups’callus regenerative capacity is better,5th,7th groups optimizing mature embryo callusregeneration system can achieve excellent regenerative capacity,6th,8th,9th groups ofmature embryo callus regeneration capacity is worst. 4. There were significant differences of Soluble protein content, PPO activity and dryweight in many groups,. PPO activity of common wheat callus can be used as predictiveindicators of tissue regenerative capacity, soluble protein can be used as an indicator topredict callus browning, moisture also can be used as an indicator to predict the rate ofbrowning.5.31resistant plantlets have survived of49Transplanting plantlets.31resistant plantshave three positive plants, There were two of Zhengmai9023,one of Xinong1376, Xiaoyan22did not get a positive plants, the conversion rate is0.17%.
