Tetrastigma hemsleyanum Diels et Gilg is a perennial sprawl vine plant belonging to vitaceae and Tetrastigma. This peculiar and rare plant is used as traditional Chinese medicine to treat children’s fever, chronic pharyngitis and tumor et. Especially in antitumor, it brings good news for cancer patients.Because the wildlife resource of Tetrastigma hemsleyanum has been over excavated and becomes in imminent danger, Therefore, Zhejiang province has list it as one of those germplasm resources that must be protected, and because of the technical obstacle in artificial cultivation, the technique of adventitious roots culture can be used to produce anti-cancer flavonoids. This will not only develop the rare-touched research field in its adventitious roots, but also meet the market demand while protecting the wildlife resources. This study used the leaves of Tetrastigma hemsleyanum as explants, designed the experiments to induce callus, proliferation, adventitious roots differentiation and proliferation, and analyzed the factors influencing these processes. We also tested the hormone conditions for adventitious roots proliferation, with the purpose to provide technical support for industrialized production of anti-cancer flavonoids from Tetrastigma hemsleyanum’s adventitious roots.The major conclusion of this study:(1) The optimal medium formula for induction of Tetrastigma hemsleyanum callus is MS+30g·L-1Sucrose+2.0mg·L-16-BA.+0.2mg·L-1NAA. Under this condition, the induction rate is100%, the callus is big, green and dense.(2) The optimal medium formula for callus sub-cultural proliferation is MS+30g· L-1Sucurose+2.0mg· L-16-BA+0.2mg·L-1NAA; the optimal temperature is28℃, using daylight lamp to save cost. To further increase the content of dry matter, the mixture of red and blue lights at a ratio of9:1or7:3can be used.(3) The optimal medium formula to induce the differentiation of adventitious roots is3/4MS/MS+1.0mg· L-1NAA, with the sucrose concentration of20~40g· L-1.(4) The optimal medium formula for adventitious roots propagation is MS+30g· L-1sucrose+1.0mg·L-1NAA+1.0mg·L-16-BA, the rate of increase in fresh weight of adventitious roots is145.56%; UV spectrometry analysis of adventitious roots upon various treatments detected flavonoids-like compounds with the highest content of24.4mg· g-1· DW-1, which is higher than the total flavonoids in the callus tissue.
