Tetrastigma hemsleyanum Diels et Gilg is a kind of medicinal herbs,of which flavonoids have anti-cancer effects.In this paper,T.hemsleyanum from four habitats(Zhejiang Province,Jiangsu Province,Hunan Province and Fujian Province)were taken as materials to study the growth characteristics,physiological,biochemical indexes,photosynthetic pigments,flavonoids enzyme activities and total flavonoids contents in different parts.Subsequently,the expression of major flavonoids and polysaccharide synthase genes was detected by quantitative PCR.(1)There were differences in growth characteristics,soluble substance and photosynthetic pigment contents of T.hemsleyanum from different areas.The colors of stems and leaves were darker in those of Jiangsu,Zhejiang and Hunan provinces.With the increase of latitude,stem diameters and leaf areas of T.hemsleyanum were smaller,soluble amino acid contents were decreased,soluble protein and soluble sugar contents both showed an increasing trend.Chlorophyll a,chlorophyll b,total chlorophyll and carotenoid contents leaves from Hunan was higher than those from other three provinces.(2)The activities of flavonoid synthase were different in diversity areas of T.hemsleyanum.Contents of PAL,CHS and CHI were respectively highest in leaves from Fujian,Cangnan and Chun’an in Zhejiang.When root tubers were used as experimental materials,the highest enzyme activities were in Quzhou(PAL),Taizhou(CHS),Longquan(CHI)of Zhejiang Province.In addition,total flavonoid contents varied with different areas,while flavonoid contents in leaves and root tubers were also different,with the highest contents in Fuyang(leaves)and Fujian(root tubers)respectively.(3)Results of quantitative PCR analysis showed expression levels of flavonoids and polysaccharide synthase genes in leaves and roots of T.hemsleyanum.There were 21 enzyme genes expressed in flavonoid synthesis pathways,and 17 of them were highly expressed in leaves than those in roots.5 polysaccharide enzyme genes in roots expressed more than those in leaves.
