Tetrastigma hemsleyanum is a vine of Tetrastigma(Vitaceae)and it is a unique and rare medicinal plant in China.The whole plant of T.hemsleyanum can be used medicinally,and its underground roots and roots have the best medicinal effect.In recent years,with the in-depth study of the pharmacological effects and the understanding of pharmacological value of T.hemsleyanum,its social needs have become more and more extensive.Under the condition of limited wild resources,the development of artificial cultivation is important to improve the yield and quality of T.hemsleyanum.The problem of low yield of roots and the content of medicinal components in the artificial cultivation needs to be solved,so it is extremely necessary to explore new cultivation techniques.This experiment was conducted at the Kuaiji Mountain Base in Pingshui Town,Shaoxing City.The growth,physiological characteristics,and active ingredient content of T.hemsleyanum were studied under different shading intensities,different forest types,and different light quality treatments.Comprehensively determine the indicators,select the best light quality,light intensity,and cultivating forest type conditions to provide a theoretical basis for the artificial cultivation of T.hemsleyanum.The specific findings are as follows:1?Using three-year-old T.hemsleyanum as material,design the light quality cultivation test of different color filters(red,yellow,green,blue,white)to study the growth traits,physiological indicators,biomass and tuber content of total flavonoids.The results showed that the leaf length,leaf width,and leaf area of T.hemsleyanum under green light treatment were the largest,while those above blue light treatment were the lowest,and T.hemsleyanum under red light and yellow light treatment was better.The contents of chlorophyll a,chlorophyll b and total chlorophyll in leaves of T.hemsleyanum were ranked as follows: white light>red light>yellow light>green light>blue light;the content of soluble protein in leaves was the highest with blue light(5.966 mg·g-1),and the lowest with red light(3.976 mg·g-1);soluble sugar content with red light and yellow light is higher,and the treatment with blue light is the lowest.The biomass of the upper and lower parts of T.hemsleyanum was the largest under the red light treatment,and the lowest under the blue light treatment.The total flavonoid content of the tuber was highest under the green(0.431%)and red light(0.456%)treatment.The yield and quality of clover tubers are better under red light treatment,so it is recommended to increase the red light treatment during the cultivation of T.hemsleyanum.2.Dynamic response values of physiological indicators and medicinal compositions of T.hemsleyanum were studied under different light intensity conditions(0,50%,70%,and 90% of natural full illumination).The results showed that the physiological indicators of T.hemsleyanum showed dynamic changes with the growth process and the processing time.With the increase of shade,the chlorophyll content gradually increased,the soluble sugar content gradually decreased,and the soluble protein content increased first and then decreased.The tuber yield and total flavonoid content increased at first and then decreased,and the tuber yield and total flavonoid content reached the highest at 70% of shading.Studies have shown that proper shading is beneficial to the growth of T.hemsleyanum and its accumulation of medicinal components,and tuber yield(32.44 g)and total flavonoid content(1.162%)reach a relatively high value at 70% shading,which is a suitable degree of illumination for cultivation.3.The variation of the growth,physiological indexes and medicinal constituents of T.hemsleyanum were studied under different conditions of light intensity(Cinnamomum camphora,Cyclocarya paliurus,and bamboo forests).The results showed that the leaf growth of T.hemsleyanum was better in C.camphora and bamboo forests;the contents of chlorophyll and soluble protein were higher in different forest types than in the control,and the higher chlorophyll in C.camphora and bamboo forest than C.paliurus from June to July.The contents of chlorophyll and soluble protein were highest in C.paliurus from August to November.The content of soluble sugar in three different types of forest was lower than that of the control.Among them,soluble sugar was the highest in C.camphora,and the lowest soluble sugar was found in bamboo forest.The yield(48.63 g)and medicinal composition(0.686%)of C.paliurus was highest in the harvest season.Therefore,it is advisable to cultivate T.hemsleyanum under the C.paliurus forest.
