| With11varieties of foxtail mille, by the some physiological indexes were evaluated drought tolerance using PEG-6000to simulating water stress method at germination stage.The varieties of drought resistance and sensitivity were selected, and identified and evaluated its drought resistance during three periods such as the stages of seed germination, seedling and mature. The effect of drought stress to millet was studied through the determination of the physiological and biochemical indexes under drought stress conditions, the analysis of changes of the seed endogenous hormones within42h of germination and the investigation of expression of key enzymes genes in Gibberellin using Real-Time fluorescence quantitative RT-PCR.The results show that:The results show that:(1) On the condition of drought in the PEG, measuring the relative germination rate, relative germination potential, relative germination index, relative plumule length and relative radicle length of the seed.The results showed that:the relative germination potential of Jingu45was the highest, while Jingu42was the lowest than other varieties. The relative germination rate of77-322was significantly lower than other varieties. The relative germination index of Jingu48,45and28were significantly higher than other varieties and Jingu42was the lowestThe sort for11varieties of drought resistance was completed using the membership function method:45>28>48>46>36>51>21>34>41>77-322>42.(2)By measuring of physiological indexes of sprout period of four different varieties of milletThe content of protein, proline and MDA, and the activity of SOD and POD increased compared with CK under drought stress.The highest accumulation of soluble protein was Jingu28and the content of proline in42and77-322were significantly lower than28and45. The accumulation of POD was the highest of Jingu28,and the lowest was42under the PEG stress. The SOD activity of28,45,42,77-322has respectively increased by266.35%,113.84%,57.40%and47.13%than CK, and the increment of content of42MDA was the highest among them.(3) We had studied the drought resistant of millet seedling,in the way of simulation of field trials in the laboratory.Through the method of direct dry of soil,it measures the physiological and biochemical index determination of the millet seedling leaf,it was showed that the content of chlorophyll and the moisture of leaf decreased under drought stress and the content of soluble protein and proline,activity of SOD,POD, CAT increased.The sorts the for four kinds of millet of drought resistance was completed using of membership function method:45>28>77-322>42.(4) It shows that the unit production of jingu28had the highest yield and the growth of Jin Gu45was better through the field experiment on agronomic traits survey in the millet seedling and mature stage.(5) Using enzyme linked immunosorbent assay (ELISA) to measure the changes of different varieties’ endogenous hormonal in seed germination period (including IAA,GA,ZR^ABA and JA) under drought stress. The studies had shown that:the content of GA, ABA gradually declined,the content of JA increased and IAA and ZR had complicate changes under PEG stress.(6) With the real-time fluorescent quantitative PCR technology, the gene expressions of seven key enzymes of gibberellins were analyzed, which showed that KAO expression of variety42was higher than45in the early seed germination, but lower in the late germination; GA2ox expression was always higher than45of strong drought resistance; the expression of GA20ox had no significant difference between the two varieties. |
