Endometritis is one of the most common major diseases in cows, which would lead to inheat disorders and infertility although repeat breeding in case of untreated or poorly treated,therefore, resulting in low breeding efficiency of cows. Severe endometritis may causesystemic infections, sepsis, even resulting in death. The productive efficiency of cattle couldbe effectively improved by preventing the occurrence of endometritis. In order to screen andoptimize the traditional Chinese herbal formula, the pharmacodynamics of the traditionalChinese herbal formula by decomposing its compounds was carried out in the studies, forproviding effective traditional Chinese medical formula in prevention and treatment of cowendometritis and researching its anti-inflammatory effect, in order to provide scientific basisfor a new veterinary drug research and development.1. The antibacterial effects of traditional Chinese medical formula and its disassembledcompounds in vitro: the bacteriostatic effect of the traditional Chinese midical formula YujinSan and its ingredients on three main pathogens of cow endometritis were studied andanalyzed by using orthogonal t value method. The results showed that the best antibacterialeffect of Yujin San and its ingredients on S. aureus were Fructus Chebulae, Radix et RhizomaRhei, Rhizoma Coptidis and Cortex Phellodendri Chinensis, the best antibacterial effect ofYujin San and its ingredients on E. coli were Fructus Chebulae, Radix Scutellariae andRhizoma Coptidis, the best antibacterial effect of Yujin San and its ingredients onStreptococcus were Radix Curcumae, Rhizoma Coptidis and Cortex Phellodendri Chinensis.According to the contribution of every herbal medicine on the three tested bacteria to the Dvalue, the Fructus Chebulae and Rhizoma Coptidis were considered to be the best ingredientsof Yunjin San for antibacterial effect. Dose optimization combination test results show that theratio of myrobalan: rhizoma coptidis=2:1is the best ratio. Its bacteriostatic effect on threemain pathogens of cow endometritis isolated from clinic was in accordance with standardstrains, which showed that the optimum combination has good bacteriostatic action on cowendometritis pathogens.2. The effect of optimized Yujin San on secretion of inflammatory cytokines from mouse macrophage: An inflammation model of mouse macrophages induced by bacteriallipopolysaccharide (LPS) was constructed in vitro, to study the effect of optimized Yujin Sanon mouse peritoneal macrophages, including the poison effect, the proliferation effect, and thephagocytosis effect, the dosage dependent effect of optimized Yujin San on secretion of nitricoxide (NO), nitric oxide synthase (NOS), tumor necrosis factor (TNF-α), and interleukin-6(IL-6). The results showed that there were not obvious toxic effects on mouse peritonealmacrophages between the concentration range of156.3~312.5μg/mL, the proliferation andphagocytosis of mouse peritoneal macrophages could be promoted in the concentration rangeof9.4~150.0μg/mL. Results also showed that a significantly inhibition effect on secretion ofpro-inflammatory cytokines, including TNF-α, IL-6, NO and NOS etc. was found inconcentration range of9.4~150.0μg/mL in a dose-dependent manner. The anti-inflammatoryeffect of optimized Yujin San in vivo: anti-inflammatory effects of optimized Yujin San in vivowere carried by xylene-induced ear edema and formalin-induced paw edema in mice. Theresults showed that effects of middle and high concentrations of optimized Yujin San onxylene-induced ear swelling is the same with dexamethasone treatment group, there was notsignificant between middle concentrations groups and high concentrations groups (P>0.05),while middle concentration was significantly better than the low concentration group(P<0.05), which showed that the inhibition effect of middle concentration is better in mice earedema induced by xylene; the inhibition of optimized Yujin San middle and highconcentration on formaldehyde-induced paw edema is the same as dexamethasone treatmentgroup (P>0.05), and the high concentration group was significantly better than the low-dosegroup (P<0.05), there was not difference between middle concentration group and lowconcentration group (P>0.05), which indicating the inhibition effect of high concentration isbetter on formaldehyde-induced paw edema.
