The production of milk was increasesed significantly in high producing dairy cows with the advance of technology and dairy farming management conditions. At the same time, nutrition metabolic disease problem was come out in dairy cows, and there is a serious threat to the production and development of modern animal husbandry industry, especially hyperketonemia and hyperlipidemia in cows, this study was focus on the relationship between cows high ketone hematic disease and liver、kidney metabolism around parturition, and the relationship between cows hyperlipidemia and leptin. For two aspects to provides the theory basis for clinical prevention and treatment of cow perinatal nutrition metabolic diseases. There were four parts in this research.Part1:Do the research of cows partial liver function index of the dynamic monitoring in plasma around calving, in order to analysis the relationship between the metabolism of liver function and high ketonemia cows and others. There were32cows were selected with similar ages, body conditions and tire in this experiment, according to ketone bodies content in plasma is greater than or less than6mg/dL for a tipping point, experimental cows can be divided into the experimental group (n=9) and the control group (n=23), respectively in the prenatal, parturition day and7,14,21,28,35,42,49,56,63days postpartum to test the cow partial liver function (NEFA, AST, GGT, DBIL, TBIL, AP, ALB, GLB). Comparing the relationship between liver function index and ketone bodies of two groups, we found that it is significant positive correlation of free fatty acids and ketone bodies in high ketone group and the control group around parturition(p<0.05), ROC curves of free fatty acids and ketone bodies shows that screening of high ketonemia cows in postpartum by free fatty acid was significance in clinical study, and the Cut-off value was754μmol/L.The correlation of ketone bodies and AST in high ketone group in postpartum was extremely significant positive(p<0.01), the correlation of ketone bodies and DBIL was significant positive(p<0.05). There was extremely significant positive correlation between concentration of ketone bodies and ALB in cows postpartum in control group (p<0.01), and there was a significant negative correlation between ketone bodies and GLB in cows postpartum in control group (p<0.05).Part2:Research on the influence of renal function indexes around parturition on hyperketonemia,32holstein cows were test in prenatal period, renal function indexes (BUN, CR, UA, LD) had been dynamic determined on7days perparturm, parturition day and7,14,21,28,35,42,49,56,63days postpartum respectively. Results show that overall mean of plasma BUN was7.21±2.24mmol/L in high ketone bodies group, the value was significantly higher than control group (6.27±2.13mmol/L). Overall mean of CR in high ketone group is82.06+20.72μmol/L, the control group was72.22+21.42μ, mol/L, it was significantly different from the value of high ketone group. There was a significant positive correlation between high ketone bodies and BUN in plasma around calving in high ketone group (p<0.05), and there was significant negative correlation in CR and LD with ketone bodies.Part3:Explore the relationship between lipid metabolism and the change of leptin concentrations around cows parturition,32holstein cows were selected in Nanning region. Study of perinatal lipid metabolism and leptin dynamic changes around calving, experimental cows were divided into hyperlipidemia group (n=8) and the control group (n=24), comparing two groups of lipid metabolism changes, we found that leptin concentrations in experimental cows was lower than control group, but no significant difference (p>0.05). It was positive correlation between leptin and triglycerides, total lipid, total cholesterol in hyperlipidemia group (p<0.05), and the same as control group. Comparing the correlation coefficient of two groups of leptin and triglyceride, correlation in hyperlipidemia group (r=0.196, p<0.1) between lipid metabolism substance and leptin concentration was slightly better than the control group (r=0.156,p<0.05).Part4:In order to do research of the influence of hyperketonemia on milk production and breeding in cows postpartum, plasma ketone body was test in this experiment in perinatal cows, base on the ketone body, experimental cows were divided into high ketone group (n=9) and the control group (n=23). Tracking and monitoring body condition scoring, milk production and breeding performance of the two groups. The result shows that estrus rate post-parturition of high ketone group was44.44%, control group was52.17%, and days of postpartum breeding in high ketone group was96.45±32.58days, calving interval was402.35±25.96days, control group was76.35±26.01days and385.46±27.02days.Conclusion:1. The activity of AST and GGT were increased to100.78U/L and29.19U/L in high ketone bodies group on the35th day after calving, these two values show that the liver of cow were damaged serious. It should be significant for the screening of hyperketonemia when the value of NEFA is754μmol/L.2. Overall mean of BUN is7.21±2.24mmol/L in high ketone bodies group, and the value of control group was6.27±2.13mmol/L. There is a significant positive correlation between BUN and ketone in high ketone bodies group.3. The research was indicated that the concentration of leptin in control group was a little bit higher than high ketone bodies group. It should be hyperketonemia of cow with decrease of leptin concentration before parturition.4. Hyperketonemia of cows can be influence on the production performance and milk yield. Cause of that milk yield of control group was increase to25.51±6.12Kg/d on the35th day after calving, but the high ketone bodies group was22.31±5.97Kg/d. And the days of first bleeding were96.45±32.58days, and the control group was76.35±26.01days.
