Objective:To investigate the protective effects of tocotrienol in mice fed with high-fat diets, and explore its regulation mechanism of lipid metabolism.Methods:(1) In vivo testing:64Kunming male mice were divided into8groups:the male mice in group1,3,5was fed with diets and group2,4,6,7,8were fed with high-fat diets, which the4,7,8group were given200,200,50,100mg/kg· bw tocotrienol separately, and group5,6were given gavage with vitamin E and tea dose100mg/kg·bw.Having fed the two months,the neck were taken off to death, the liver were acquired,the viscera index was analysized and histopathology was examined;superoxide dismutase (SOD),lipid peroxide malonic aldehyde (MDA)glutathione peroxidase (GSH-PX), total antioxidant capacity (T-AOC),catalase (CAT) of liver homogenates were detected,the RNA of liver tissue were extracted, by Tocotrienols had the effects on the expression levels of RXR-a, PPAR-a, PPAR-y were analyzed by RT-PCR and semi-quantitative PCR.(2) in vitro:L02cell line was selected as a fat model in vitro to co-culture with palmitic acid (PA).Then,the role of gamma-tocotrienol on regulating RXR-a, PPAR-a, PPAR-y was detected by RT-PCR.Results:Tocotrienols reduced the fat mice liver index, the detection of liver tissue homogenate showed that it reduced MDA content of liver on mice in fat diet group and slightly increased the level of T-AOC, a certain extent decreased the level of SOD, GSH-PX, CAT,performing the certain capacity of antioxidant and reducing the oxidative damage in the liver of mice. RT-PCR results showed that tocotrienol can enhance the expression of RXR, PPAR-y in liver, in L02cells of high-fat, gamma-tocotrienol also can improve the expression of RXR, PPAR-γ,the results in vitro and in vivo are authenticate to each other.Conclusion:(1) tocotrienols have antioxidant effects, and make a certain role in regulating lipid metabolism in the liver, and can upregulate theamount of expression of RXR-alpha, PPAR-alpha, PPAR-y, regulate lipid metabolism.(2),in the hyperlipidemia model of L02cells gamma-tocotrienol can improve the amount of expression of the RXR-alpha, PPAR-alpha, PPAR-gamma, and regulate lipid metabolism.
