In this study,healthy swine’s spleen were used as raw materials, respectively, usingconventional dialysis and ultrafiltration production each6batch of transfer factor. Theresults showed that compared to dialysis, ultrafiltration method can be better applied tolarge-scale industrial production.This test made ultrafiltration method in the production of swine’s spleen TF inspectionprocedures, to guarantee the quality of swine’s spleen transfer factor,standardize theproduction process, finally establishing a related API standards.swine’s spleen TF appearedpale yellow, clear and transparent, with special smell. pH value of6.5.Heavy metals,arsenic, chloride salt test results showed that the product color than standard shallow,proved the content within the control range.Protein detection was negative, indicating thatthe main ingredient of swine’s spleen TF was polypeptide and ribose,content ofpolypeptide were more than8.53mg/ml while ribose above218.34μg/ml. Analysis by UV,swine’s spleen TF peak at the wavelength of250-260nm.Macromolecular materialproportion is less than0.5%, free amino acid content is greater than4mg/ml.According to the China biological products procedures requirements of the swine’sspleen TF sterile testing, allergy testing, security testing, heat testing, the results showedno abnormality. By E-rosette test, mouse macrophage phagocytic function test, the swine’sspleen TF activity was measured. Results show that in the E-rosette test, swine’s spleen TFcan make the number of rosette formation rate were significantly improved. In the mousemacrophage phagocytic function test, swine’s spleen TF could significantly improve thephagocytic index and phagocytic rate. Sub block test immune mouse model by swine’sspleen transfer factor solution and the chromatography group, the result shows that theimmune dope and component1of the normal mice inhibited model can enhance theimmune function.
