Sedum spectabile Bareau is a perennial flower of sedum genus of sedum family, is an important landscaping material, It is important landscaping material with good viewing, mostly based division propagation or cutting propagation. And it’s rare sowing, extensive management and resistants to cold and other harsh environments, diseases and pests. Sedum spectabile Bareau there are shortcomings, for example, their application in landscape plant type, single color and short flowering period. In this study, we use Sedum spectabile Bareau as test material, give Sedum spectabile Bareau mutagenic treatment with different concentrations of chemical mutagens (EMS) and different radiation dose60Co-y rays. Through the statistics of the germination rate of generation after mutagenic treatment, determine and analysis plant morphology index, measuring physiological indicators, observe leaf tissue structure, Study on half death and appropriate mutagenic concentration and radiation dose, initial screening mutant lines with high ornamental value, in order to provide the theoretical foundation and rich breeding material for Sedum spectabile Bareau. The main results of this test are as follows:1. The LC50of Sedum spectabile Bareau seeds by EMS treatment is0.7%; and the suitable concentration for mutagenesis is0.5%to0.7%. The LD50of Sedum spectabile Bareau seeds by60Co-y radiation treatment is70Gy; and the suitable radiation for mutagenesis is50Gy to70Gy.2. After the physical and chemical mutation treatment of Sedum spectabile Bareau seeds, the leaf type, plant type, flower type varied traits. Leaf type of EMS treatment groups with asymmetry, crimping phenomenon; the variances of plant are mainly low plant and gather plant; the variances of flowers are mainly for large flower color deep, rich multicolored decorations. Leaf type of60Co-γ radiation treatment groups with lanceolate leaf, curling leaf; the variances of plant are mainly low strain, gather plant and flat type;60Co-y radiation associated variations of flowers arising out of the performance of color depth, large diameter for flowers, flowers dense.3. Physical and chemical mutagenesis significantly affect stoma morphology and organization structure of leaves of Sedum spectabile Bareau, Stoma aperture of EMS treatment groups are smaller than the control group, and0.9%treatment group mutant leaf stoma aperture is the smallest; EMS treatment groups palisade tissue/spongy tissue ratios are different from the control group significantly, In addition to the1.3%treatment group, other treatment groups are higher than the control group,and the0.5%treated group is the highest, can be as high as0.847; The ratio of palisade tissue/leaf thickness is significantly higher,,and the0.9%treated group is the highest, can be as high as0.389.60Co-γ radiation treatments stoma aperture variation of leaves vary in sizes, and70Gy treatment group mutant leaf stoma aperture is the smallest,200Gy treatment group mutant leaf stoma aperture is the largest; larger than the control group; the higest group of the ratio of palisade tissue/spongy tissue is70Gy treatment group, can be as high as0.831, The ratio of palisade tissue/leaf thickness is significantly higher, and the70Gy treated group is the highest, can be as high as0.345.4. Physical and chemical mutagenesis have impact on Sedum spectabile Bareau physiological indicators. Chlorophyll content was negatively correlated with the concentration of EMS; The active of POD、CAT increasing with the concentrations of EMS showed a downward trend after the first rising; The active of SOD increased at0.9%concentration then began to decline after the peak, at1.1%concentration treatment group followed a modest rise of1.3%treatment group decreased until SOD activity reached the lowest; Soluble sugar, soluble protein content with increasing concentrations of EMS first decreased and then increased; The content of MD A increased with the concentrations of EMS. Chlorophyll content and60Co-y radiation dose was negatively correlated; The active of POD、CAT with the increasing of radiation dose showed a downward trend after the first rising; SOD activity began to decline after50Gy active treatment group reached the peak,150Gy-200Gy a slight rise slightly; Soluble sugar, soluble protein content increasing with60Co-y radiation dose increased and then decreased after the first show; The content of MDA has upward trend with the increase of60Co-y radiation dose.5. In this study, we obtain10variant strains with high ornamental value, among which there are5variant strains having high ornamental value in plant type and leaves. Including strain of gathering up plant type and small entire blade; strain of thick blade, gathering plant type, curly upwards blade; strain of low plant, shortening internode, curly and downward blade; strain of low plant, shortening internode, lanceolate curly and downward blade and dense blade; strain of gathering plant type, curly upwards blade; strain of mainly flat plant type, leaf tips curly upwards slightly. There are5strains with high ornamental value in flowers among the mutants. The main expression is that thick inflorescence branch, large diameter of inflorescence size and flower, bright beautiful color, compact and dense flowers.
