With the development of life sciences, the study of life has gone deep into theinternal details. And two-photon fluorescence microscopy enables biologicalspecimens imaging in three dimensions with sub-micrometer resolution. However,there is no general software system to realize the imaging of two-photonfluorescence microscopy. So based on the two-photon fluorescence microscopy, thepaper designed and implemented a software system to collect the image data, topre-process the image information and to reconstruct image in the LabVIEW andVS2008with OpenGL software environment. The contribution of this paper isorganized as follows.Firstly, taking comprehensive system functional requirements intoconsideration, the paper designed the hardware connection and the softwareprogramming of system, and the system was divided into controller module,mechanical scanning module, image acquisition and reconstruction module andthree-dimensional reconstruction module.Secondly, in mechanical scanning module, three different feasible methods ofscanning are proposed. Two scanning paths, rectangle path and spiral path, weredesigned. Only the rectangle path was used for scanning in this system, because itcould collect the grid data to reconstruct image directly.Then, in the process of image acquisition and reconstruction, the image datawas collected at the same time as scanning. This method improved thesignal-to-noise level. An ideal sampling frequency was selected to ensure imageacquisition without image distortion.Finally, in the process of three-dimensional reconstruction, ray castingalgorithm was applied. The process of3D reconstruction in our system consisted ofthe collection, reconstruction and visualization of volume data. LabVIEW softwareplatform was used to realize collection, mean filtering and DW algorithm basedreconstruction of the volume data. VS2008software platform combined withOpenGL graphics software interface was used to render the3D image.
